目的探讨P物质(substance P, SP)及其受体神经激肽-1(neurokinin-1 receptor, NK-1R)在鼻咽癌组织中的表达及意义。方法收集鼻咽癌患者的肿瘤组织及鼻中隔偏曲患者的正常鼻咽黏膜组织,采用实时荧光定量PCR和Western blot检测SP/NK-1R mRNA和蛋白表达水平;免疫组织化学法观察SP/NK-1R表达与分布规律,分析SP/NK-1R表达与鼻咽癌患者临床参数之间的相关性;CCK-8和Transwell检测SP对鼻咽癌HNE1细胞株增殖、侵袭的影响。结果SP和NK-1R mRNA在鼻咽癌组织中相对表达量(7.96±1.75, 7.12±1.54)比正常鼻咽黏膜组织(1.05±0.34, 1.04±0.28)明显增高(P<0.01);SP和NK-1R蛋白在鼻咽癌组织中相对表达量(1.18±0.27, 0.81±0.17)比正常鼻咽黏膜组织(0.56±0.06, 0.48±0.05)明显增高(P<0.01);SP和NK-1R在鼻咽癌组织阳性表达率高于正常鼻咽黏膜组织(64% vs 11%, 68% vs 22%,P<0.01),并且SP和NK-1R在TNM分期Ⅲ/Ⅳ期患者肿瘤组织中的阳性表达率高于Ⅰ~Ⅱ期患者(P<0.05);SP(10-9~10-7 mol/L)可诱导HNE1细胞增殖(P<0.05)、侵袭(P<0.01)。结论SP/NK-1R在鼻咽癌组织中高表达并与鼻咽癌TNM分期具有相关性,有望成为鼻咽癌新的诊治靶点。

Abstract:

ObjectiveTo investigate the expression of substance P (SP) and its receptor, neurokinin-1 receptor (NK-1R), in nasopharyngeal carcinoma (NPC) tissues and their correlation with the clinicopathological parameters of the patients. MethodsReal-time fluorescence quantitative PCR (RT-qPCR) and Western blotting were used to detect the expression of SP and NK-1R mRNAs in the specimen of tumor tissues of NPC patients and normal tissues from patients with deviated nasal septum, and their protein expression and distribution in the tissues were observed using immunohistochemistry. Chi-square test was used to analyze the correlation of the expression of SP and NK-1R with the clinicopathological characteristics of the patients. CCK8 and Transwell assays were used to evaluate the effects of SP on the proliferation and invasion of HNE1 cells. ResultsThe mRNA levels of SP and NK-1R were significantly increased in NPC tissues (7.96±1.75 and 7.12±1.54, respectively) as compared with the normal tissues (1.05±0.34, 1.04±0.2; P<0.01); the protein expression of SP and NK-1R were also significantly upregulated in the NPC tissues (1.18±0.27 and 0.81±0.17, respectively) in comparison with the normal tissues (0.56±0.06, 0.48±0.05; P<0.01). The positive expression rate of SP and NK-1R in the NPC tissues was 64% and 68%, respectively, significantly higher than the rates in normal tissues (11% and 22%, respectively; P<0.01). The positive expression rates of SP and NK-1R were significantly higher in stage Ⅲ and stage Ⅳ NPC cases than in stage Ⅰ-Ⅱ cases (P<0.05). Exogenous SP significantly enhanced the proliferation (P<0.05) and invasion (P<0.01) of HNE1 cells in vitro. ConclusionSP and NK-1R are over-expressed in NPC tissues, and their expression levels are correlated with the TNM stages of NPC, suggesting their value as potential new targets for diagnosis and treatment of NPC.
