目的研究长链非编码RNA LINC02163在肝细胞肝癌(hepatocellular carcinoma,HCC)组织及肝癌细胞系中的表达及对肝癌细胞迁移、侵袭的影响。方法利用Quantitative Real-time PCR(qRT-PCR)检测LINC02163在5株肝癌细胞系(HuH-7、HCCLM3、MHCC-97H、SNU-182和SMMC-7721)、正常肝细胞系(L02)和60例肝癌组织及其配对癌旁组织中的差异表达情况,分析LINC02163的表达水平与肝癌患者临床病理参数的关系;利用LipofectamineTM 2000转染LINC02163小干扰RNA(siRNA)至HuH-7、HCCLM3肝癌细胞内敲低LINC02163表达;通过Transwell实验检测敲低LINC02163后肝癌细胞迁移、侵袭能力的改变。结果LINC02163在肝癌组织中的表达水平明显高于配对癌旁组织(P<0.001);LINC02163在肝癌组织中的表达与TNM分期、微血管侵犯(MVI)明显相关(P<0.01);与正常肝细胞系(L02)比较,LINC02163在5株肝癌细胞系中高表达(P<0.001),其中在HuH-7细胞系中表达最高,其次是HCCLM3细胞系;肝癌细胞HuH-7、HCCLM3敲低LINC02163的表达后迁移和侵袭能力明显降低(P<0.01)。结论LINC02163在肝癌组织和肝癌细胞系中的表达上调,其可促进肝癌细胞的迁移和侵袭,故可能与肝癌的发生、发展密切相关。

Abstract:

ObjectiveTo investigate the expression of long non-coding RNA LINC02163 in hepatocellular carcinoma (HCC) tissues and HCC cell lines, and its effect on the migration and invasion of hepatocellular carcinoma cells.  MethodsThe differential expression level of LINC02163 in 5 HCC cell lines (HuH-7, HCCLM3, MHCC-97H, SNU-182 and SMMC-7721), normal liver cell line (L02) and 60 HCC tissues and paired adjacent tissues was determined by quantitative real-time PCR (qRT-PCR), and the relationship between its expression level and the clinicopathological parameters of HCC patients was analyzed. LipofectamineTM 2000 was used to transfect LINC02163 small interfering RNA (siRNA) into HCC HuH-7 and HCCLM3 cells in order to knockdown the expression. Then the changes of migration and invasion ability of HCC cells after the knock-down of LINC02163 were examined by Transwell assay. ResultsThe expression level of LINC02163 in HCC tissues was significantly higher than that in paired adjacent tissues (P<0.001), and the level was significantly correlated with TNM stage and microvascular invasion (MVI) in the patients (all P<0.01). Compared with normal liver cell line (L02), LINC02163 was highly expressed in 5 HCC cell lines (P<0.001), among which HuH-7 cells had the highest expression level, followed by HCCLM3 cells. LINC02163 knock-down significantly reduced the migration and invasion abilities in HuH-7 and HCCLM3 cells (P<0.01).  ConclusionLINC02163 is up-regulated in HCC tissues and cell lines and promotes the migration and invasion of HCC cells, which may be closely associated with the occurrence and development of HCC.
