摘要:

目的 探讨LINC00475在不同WHO分级胶质瘤标本中的表达变化,并在胶质瘤细胞系LN382 U87MG中验证LINC00475对增殖、迁移、侵袭功能的影响。方法 利用TCGA胶质瘤数据库对LINC00475进行差异表达分析、ROC曲线分析和生存分析。采用qRT-PCR技术检测LINC00475在本院临床搜集的胶质瘤样本中的表达,分析LINC00475是否存在差异表达以及与肿瘤分期分级有无相关性,与患者预后是否相关。分别在LN382和U87MG细胞系上采用RTCA实验检测NC组(阴性对照)和silence组(敲减LINC00475)增殖、侵袭、迁移功能的改变。结果 TCGA数据库提示LINC00475在肿瘤组织中表达显著上调(P<0.001),且ROC曲线下面积等于0.98,与患者预后显著负相关(P<0.01)。qRT-PCR结果证实LINC00475在胶质瘤组织(2.62±0.89 )中的表达显著高于正常组织(1.00±0.36),且与肿瘤分期、分级呈正相关(WHO Ⅱ: 1.60±0.40,WHO Ⅲ:2.65±0.63, WHO Ⅳ:3.30±0.58)。LINC00475高表达组患者总体生存率显著低于低表达组(P<0.05)。两个细胞系的silence组增殖、侵袭、迁移能力显著低于NC组(P<0.01)。结论 LINC00475表达在胶质瘤组织中显著上调,且和WHO分级正相关;高表达的LINC00475促进了胶质瘤细胞增殖、侵袭、迁移能力。

Abstract:

Objective To investigate the differential expression of LINC00475 in brain tissues at different WHO grades of glioma, and determine the effect of LINC00475 on the proliferation, migration, and invasion of glioblastoma cell lines LN382 and U87MG. Methods With TCGA-GBM database as the analytical foundation, differential expression analysis, ROC curve plotting, and survival analysis were performed towards LINC00475. The expression of LINC00475 in normal brain tissues from 10 patients with brain trauma and glioma tissues of 48 patients was detected with qRT-PCR. The relationships of differential expression of LINC00475 with WHO grades and with prognosis were analyzed. Real time cellular analysis (RTCA) was applied to detect the cell proliferation, invasion and migration between the negative control cells and the cells after LINC00475 knockdown in LN382 and U87MG cells. Results The analysis of TCGAGBM database indicated that the expression of LINC00475 was significantly up-regulated in glioma tissues (P<0.001), the area under the ROC curve equaled to 0.98, and there was a remarkably negative correlation between LINC00475 expression and patient prognosis (P<0.01). qRT-PCR results demonstrated a much higher level of LINC00475 in the glioma tissues than normal tissues (2.62±0.89 vs 1.00±0.36), and the expression level was positively correlated with WHO grades (WHO Ⅱ: 1.60±0.40, WHO Ⅲ: 2.65±0.63, WHO Ⅳ: 3.30±0.58). Besides, the patients with high LINC00475 expression possessed significantly lower overall survival rate than those with low expression (P<0.05). Moreover, the capacities of cell proliferation, invasion and migration were notably decreased in the silenced cells than the normal cells in on both cell lines (P<0.01). Conclusion LINC00475 is obviously up-regulated in glioma tissue, and is positively correlated with WHO grades. Its higher expression improves the proliferation, invasion and migration capacities in the tumor cells.
