摘要:

目的     研究lncRNA MUC5B-AS1基因在肺腺癌中的表达情况及对肺腺癌细胞系生物学行为的影响。方法    利用RT-PCR、qRT-PCR检测肺腺癌细胞系中MUC5B-AS1基因的表达情况;构建 MUC5B-AS1过表达载体,通过转染构建 MUC5B-AS1过表达细胞系;采用CCK-8检测肺腺癌细胞的增殖情况;应用Transwell小室检测MUC5B-AS1过表达对肺腺癌细胞系迁移和侵袭的影响。结果     检测了MUC5B-AS1在4株肺腺癌细胞系(A549、SPCA1、H1975和H1299)和1株肺正常上皮细胞系(HBE) 中的表达情况,发现MUC5B-AS1在H1299细胞系中表达最低,同时A549细胞系表达最高;通过转染构建 MUC5B-AS1过表达H1299、A549细胞系,利用qRT-PCR检测细胞系中MUC5B-AS1基因的表达,与对照组比较,MUC5B-AS1基因在H1299、A549细胞系均升高,且有统计学差异(P<0.05);CCK-8检测细胞增殖,与对照组比较过表达MUC5B-AS1对H1299、A549细胞生长并无影响(P>0.05);应用Transwell小室检测MUC5B-AS1过表达对肺腺癌细胞系迁移和侵袭,过表达MUC5B-AS1促进H1299、A549细胞的迁移和侵袭,差异有统计学意义 (P<0.05)。结论     MUC5B-AS1可能在肺癌的迁移和侵袭发挥重要作用,但对肺腺癌细胞的增殖能力没有显著影响。

Abstract:

Objective    To investigate the expression profiles of lncRNA MUC5B-AS1 in lung adenocarcinoma cell lines and the effect of MUC5B-AS1 on the biological behaviors of the lung adenocarcinoma cells. Methods     The expression level of MUC5B-AS1 in 4 lung adenocarcinoma cell lines (H1299, H1975, SPCA1 and A549) and a normal lung epithelial cell line (HBE) was detected by reverse transcription PCR (RT-PCR) and real-time quantitative PCR (qRT-PCR). LncRNA MUC5B-AS1 was overexpressed by transfection of overexpression vector in H1299 and A549 cells. Cell counting kit-8 (CCK-8) assays were performed to measure the proliferation of lung adenocarcinoma cell lines. Transwell chamber assay was performed to observe the cell invasion and migration ability in the MUC5B-AS1 overexpression H1299 and A549 cells. Results     MUC5B-AS1 was expressed in 4 strains of lung adenocarcinoma cell lines and normal lung epithelial cell line. Its expression level was quite lower in the H1299 cells, but higher in the A549 cells than other cell lines. After transfection of overexpression vector in the H1299 and A549 cells, the expression of MUC5B-AS1 was remarkably enhanced (P<0.05). CCK-8 assays indicated that MUC5B-AS1 overexpression had no effect on the proliferation in H1299 and A549 cells when compared with non-transfected cells (P>0.05). Transwell chamber assay showed the MUC5B-AS1 overexpression promoted the migration and invasion in the H1299 and A549 cells (P<005). Conclusion     MUC5B-AS1 may play an important roles in the migration and invasion in lung adenocarcinoma cells, but had no obvious effect on proliferation.
