摘要:

目的      观察传统中药成分姜黄素对葡聚糖硫酸钠(dextran sulphate sodium,DSS)诱导的小鼠溃疡性结肠炎的炎症状况及IEL亚群的影响,探讨姜黄素对治疗溃疡性结肠炎的可能机制。      方法      选取成年雄性C57BL/6小鼠60只,按照随机数字表法分为3组,每组20只,分别为正常组、DSS组和DSS+姜黄素组,通过对小鼠疾病活动程度指数(disease activity index, DAI)评分及结肠组织HE染色评估炎症状况,应用流式细胞术分析各组结肠IEL亚群。      结果      与正常组对比,DSS组DAI评分明显升高(P<0.01);结肠黏膜损伤程度明显加重;CD8αα和TCRγδ亚群比例降低(P<0.01),CD4亚群比例升高(P<0.05)。而与DSS组比较,DSS+姜黄素组DAI评分明显降低(P<0.05);结肠黏膜损伤程度明显减轻;CD8αα和TCRγδ亚群比例升高(P<0.01);CD4亚群比例降低(P<0.05)。      结论      姜黄素缓解DSS诱导的小鼠溃疡性结肠炎可能通过调节IEL亚群变化。

Abstract:

Objective      To observe the effect of traditional Chinese medicine component curcumin on intestinal intraepithelial lymphocytes (IEL) subsets and dextran sulfate sodium (DSS)-induced colonic inflammation in ulcerative colitis mice, and investigate the potential therapeutic mechanism of curcumin in inflammatory bowel disease.       Methods      Sixty adult male C57BL/6 mice were randomly divided into 3 groups: normal group, DSS group and curcumin treatment group. Disease activity index (DAI) score and HE staining were used to evaluate the inflammatory conditions. The change of IEL subsets was analyzed by flow cytometry.       Results      Compared with the normal group, the DSS group showed higher DAI score (P<0.01), lower proportion of CD8αα subset (P<0.01) and TCRγδ subset (P<0.01), and higher proportion of CD4 subset (P<0.05). Interestingly, the curcumin treatment group showed lower DAI score (P<0.01), higher proportion of CD8αα subset (P<0.01) and TCRγδ subset (P<0.01), and lower proportion of CD4 subset (P<0.05) compared with the DSS group.       Conclusion      Curcumin attenuates DSS-induced ulcerative colitis through regulating IEL subsets in the mice.
