摘要:

目的      超声引导下细针穿刺大鼠前列腺注射大肠埃希杆菌制作慢性细菌性前列腺炎模型。      方法      选取成年雄性SD大鼠36只,清洁级,体质量280~300 g,采用抽签法分为空白对照组、传统慢性细菌性前列腺炎模型(tradition chronic bacterial prostatitis model,TCBP)组和超声引导慢性细菌性前列腺炎模型(ultrasound chronic bacterial prostatitis model,UCBP)组,每组12只。4周后通过光学显微镜直接观察比较每组大鼠前列腺组织病理变化,并通过统计学方法比较炎症评分、大鼠死亡率及制作模型的手术时间。      结果      TCBP组炎症评分(腺体形态改变、炎性细胞浸润和纤维组织增生)分别为(3.22±0.31)、(3.71±0.37)、(3.80±0.29),UCBP组炎症评分分别为(3.24±0.36)、(3.65±0.41)、(3.70±0.33),均显著高于空白对照组(0.20±0.07、0.23±0.04、0.28±0.06,P<0.05),且UCBP组大鼠死亡率及手术时间均低于TCBP组(P<0.05)。      结论      超声引导下细针穿刺大鼠前列腺注射大肠埃希菌可用于制作稳定的大鼠慢性细菌性前列腺炎模型,与传统的建模方式相比具有方便、快捷、大鼠死亡率低的优点。

Abstract:

Objective        To establish a model of chronic bacterial prostatitis (CBP) in rats by fine needle injection of E. coli under the guidance of ultrasound.       Methods        A total of 36 clean adult male rats weighing 280~300 g were randomly divided into 3 groups (n=12), that is, normal control, conventional CBP model group and fine needle injection model group. In 4 weeks after model establishment, the rats were all sacrificed and their prostate was harvested for morphological observation. The inflammation score, rat mortality and operation time for making model were studied and compared.       Results        The inflammation scores in gland morphological change, inflammatory cells infiltration, and fibrous hyperplasia were 3.22±0.31, 3.71±0.37 and 3.80±0.29 respectively in the conventional CBP model group, and 3.24±0.36, 3.65±0.41 and 3.70±0.33 respectively in the fine needle injection model group. The scores of the both groups were significantly higher than those in the normal control group (0.20±0.07, 0.23±0.04 and 0.28±0.06, P<0.05). What’s more, the rat mortality was obviously lower, and  the operation time for making model was remarkably shorter in the fine needle injection model group than the conventional CBP model group (P<0.05).       Conclusion        Ultrasound-guided fine needle injection of E. coli into the rat prostate is a good method to establish stable rat CBP model, and has the advantages of easiness, convenience and lower mortality when compared with the conventional model establishment.
