摘要:

目的      探索自噬(autophagy)在小鼠实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis, EAE)病程中的时序变化,并初步探讨其在EAE中的抗炎作用及可能机制。      方法      20只C57BL/6雌性小鼠建模后分别于3、7、16、30 d用Western blot检测中枢神经系统中LC3B的变化。另将60只小鼠分为正常对照组、EAE模型组、EAE+3甲基腺嘌呤(3-Methyladenine,3-MA)组。神经功能评分观察各组小鼠神经功能变化。免疫组化检测各组小鼠免疫后7 d小胶质细胞激活情况,Western blot检测免疫后16、30 d白介素-1β(IL-1β)的表达,HE染色观察中枢神经系统炎性细胞浸润情况。      结果      ①EAE+3-MA组小鼠神经功能缺损症状较其他组严重(P<0.05)。②EAE小鼠LC3-Ⅱ/LC3-Ⅰ自免疫后7 d开始降低(P<0.05),此后一直呈低表达水平(P<0.05)。③EAE免疫后7 d即有小胶质细胞激活,EAE+3-MA组EAE小鼠小胶质细胞激活程度较EAE模型组严重(P<0.05)。④HE染色结果显示,EAE+3-MA组炎细胞浸润比同时间点EAE模型组加重(P<0.05)。⑤免疫后16、30 d,EAE模型组高表达IL-1β,EAE+3-MA组小鼠IL-1β表达水平较同时间点EAE模型组高(P<0.05)。      结论      EAE中存在着自噬水平降低,可能是导致疾病炎症反应发生的重要原因。

Abstract:

Objective      To explore the temporal variation of autophagy in C57BL/6 mice with experimental autoimmune encephalomyelitis (EAE) after immunization, and demonstrate its anti-inflammatory effect in EAE mice.       Methods      EAE models were established in 20 female C57BL/6 mice, and then the mice were killed in 3, 7, 16 and 30 d later for the expression of LC3B in the central nervous system (CNS) by Western blotting. Another 60 female C57BL/6 mice were randomly divided into the normal control group, EAE group and EAE+3-methyladenine (3-MA) group. Neurological function scores were performed to evaluate the neurological function. Immunohistochemical assay was performed to test the activation of microglia on the 7th day post immunization. Western blotting was employed to detect the expression of IL-1β in each group in 16 and 30 d after immunization. HE staining was used to observe the infiltration of inflammatory cells in the brain.        Results      ① The neural function was more seriously damaged in the EAE+3-MA group than the EAE group (P<0.05). ② Western blot results showed the expression of LC3-II/LC-3I was decreased since 7 d post immunization (P<0.05), and then kept in a lower level compared to the normal group (P<0.05). ③ There were activated microglia in 7 d post immunization (P<0.05), with more strongly activated cells in EAE+3-MA group (P<0.05). ④ There were more infiltrated inflammatory cells in the EAE+3-MA group than the EAE group at the same time point (P<0.05). ⑤ The expression of IL-1β was elevated in 16 and 30 d in the EAE group, which were more significantly in the EAE+3-MA group (P<0.05).       Conclusion      Autophagy is decreased in the EAE mice, which may be the main cause for the pathogenesis of inflammation.
