摘要:
目的      从乙型流感流行株中筛选获得可在鼠肺中稳定复制的毒株。      方法       利用免疫抑制剂甘露聚糖分别联合5株乙型流感临床分离株和1株标准株感染BALB/c小鼠,筛选出可稳定在鼠肺中连续感染传代的毒株,并纯化后感染小鼠,检测其致病力与致死率,最后对病毒核酸进行测序,分析分子特征。      结果       从5株临床分离株中筛选到1株乙型流感病毒,可在小鼠中稳定感染并致死,经测序比对发现适应过程中该株病毒血凝素(HA)发生2个突变:N212Y和E361K,神经氨酸酶(NA)发生1个位点突变S339F,其中HA E361K是裂解位点。      结论       成功从临床乙型流感分离株筛选适应获得一株鼠肺适应株,为进一步开展药物和发病机制研究提供了可能。
Abstract:
Objective        To screen a stable mouse-lung adapted strain of influenza B virus isolated from clinical obtained strains in recent years.       Methods       The influenza B viruses were isolated from clinical patients with influenza B virus only. A mouse-adapted variant of influenza B virus was obtained by 7 sequential mouse-lung passages (inoculated with mannan) and was subsequently cloned by 2 sequential plaque purifications in MDCK cells. Mouse-adapted virus was then confirmed by infection in mice and sequencing for its molecular features.       Results       A stain of mouse-adapted influenza B virus was isolated from 5 clinical strains, and able to infect and kill mice without mannan. Sequencing indicated that there were 2 mutations in hemagglutinin (N212Y and E361K), and 1 in neuraminidase (S339F). Hemagglutinin E361K was identified as cleavage site.       Conclusion        A mouse-adapted influenza B virus is obtained from adaptation of clinical viruses in mouse, and it might be as a tool for mouse infected model.
