目的      探讨PCDH10(protocadherin 10)基因诱导多发性骨髓瘤KM3细胞凋亡的作用及相关机制。      方法      采用脂质体法将重组质粒pcDNA3.1(+)PCDH10以及pcDNA3.1(+)空质粒稳定转染入人骨髓瘤KM3细胞,以未转染组和转染pcDNA3.1(+)空质粒组作为对照,通过RT-PCR和Western blot检测PCDH10基因和蛋白的表达,应用MTT法检测PCDH10转染后细胞增殖能力;应用流式细胞仪检测细胞凋亡率;应用透射电镜观察细胞凋亡超微结构;应用Western blot检测凋亡相关蛋白表达水平。      结果      PCDH10转染KM3细胞后,PCDH10基因和蛋白表达水平均明显升高。MTT结果显示PCDH10转染后细胞增殖明显受到抑制(P<0.05);流式细胞仪结果显示转染PCDH10基因的KM3细胞凋亡率明显增高(P<0.05);透射电镜下观察,PCDH10转染组细胞可以看到胞质空泡样变,核固缩以及明显的凋亡小体,对照组未见明显异常;Western blot结果显示PCDH10能降低Bcl-2的表达,促进P53、Fas的表达(P<0.05)。      结论      PCDH10基因有促进多发性骨髓瘤细胞KM3凋亡作用。
Abstract:
Objective        To determine the apoptosis-inducing effect of protocadherin 10 gene (PCDH10) on multiple myeloma (MM) cells and its mechanisms.       Methods        The human MM cell line KM3 was stably transfected with pcDNA3.1(+)PCDH10 or pcDNA3.1(+) by lipofectamine. Untransfected and pcDNA3.1(+) plasmid transfected cells were used as control. The expression of PCDH10 at mRNA and protein levels in the PCDH10-transfected cells were detected by real-time PCR and Western blotting. Cell proliferation after transfection was detected by MTT assay. The apoptotic rate was assayed by flow cytometry, and transmission electron microscopy (TEM) was employed to observe the ultra-structure of the cells. The expressions of apoptosis-related proteins were detected by Western blotting.       Results        Transfection of PCDH10 resulted in obviously increased expression of PCDH10 at mRNA and protein levels. MTT assay showed that the proliferation of PCDH10-KM3 cells was significantly suppressed (P<0.05). Flow cytometry revealed increased apoptosis in KM3 cells transfected with PCDH10 (P<0.05). TEM displayed that the PCDH10-transfected KM3 cells had cytoplasmic cavities, karyopyknosis and obvious apoptotic bodies. But no such change was found in the control cells. At the same time PCDH10-transfected KM3 cells resulted in down-regulation of Bcl-2 (P<0.05), and up-regulation of p53 and Fas gene (P<0.05).       Conclusion       PCDH10 plays a promoting role in the apoptosis of MM cell line KM3.
