摘要:
目的      探讨养血清脑颗粒对大鼠脑缺血再灌注损伤后轴突再生的影响及可能的机制。      方法      成年雄性Sprague-Dawley (SD)大鼠48只,随机分为假手术组、缺血/再灌注组、生理盐水对照组和养血清脑颗粒治疗组。缺血2周后,采用免疫组织化学及Western blot法检测缺血侧脑皮质及海马排斥性导向因子A(repulsive guidance molecular A,RGMa)蛋白的表达,免疫染色神经微丝蛋白200(NF-200)评估轴突生长。术后2、7、14 d和28 d,采用改良的神经功能缺损评分(modified neurological severity score,mNSS)评估大鼠神经功能。      结果      同缺血/再灌注组相比,养血清脑颗粒治疗组大鼠缺血侧脑皮质和海马RGMa蛋白表达明显降低(P<0.01),NF-200蛋白表达增加(P<0.05),mNSS神经功能评分显著下降(P<0.05)。      结论      养血清脑颗粒能促进大鼠缺血再灌注损伤后神经功能的恢复,其机制可能与下调RGMa蛋白的表达,促进轴突再生有关。
Abstract:
Objective      To determine the effect of Yangxue Qingnao Granule, a drug based on traditional Chinese medicine, on axonal regeneration in rats after focal cerebral ischemia reperfusion and investigate its potential  mechanism.       Methods      Forty-eight adult male Sprague-Dawley rats (SD) were randomly divided into sham-operation group, cerebral ischemia/reperfusion group, vehicle group and Yangxue Qingnao Granule treatment group. After the establishment of cerebral ischemia/reperfusion, Yangxue Qingnao Granule was given the rats at 2 g/kg (i.g.), once per day for 2 weeks. The expression of repulsive guidance molecular A (RGMa) was detected by immunohistochemical assay and Western blotting. NF-200 immunohistochemical staining was used to evaluate axonal regeneration. Neurological function recovery was evaluated by modified neurological severity score (mNSS) at 2, 7, 14 and 28 d after ischemic reperfusion.       Results      The expression of RGMa at protein level in ischemic cortex and hippocampus was significantly lower (P<0.01), but that of NF-200 was increased (P<0.05) in treatment group than cerebral ischemia/reperfusion group. The rats in Yangxue Qingnao Granule group exhibited a better neurological function recovery (P<0.05).       Conculsion      Yangxue Qingnao Granule improves neurological function recovery after focal cerebral ischemia reperfusion in rats, which may be due to its down-regulating RGMa and promoting axonal regeneration.
