摘要:
目的      探讨草麻黄中补体抑制单体(complement inhibiting component in Ephedra sinica, CICES)对大鼠脑冲击伤的保护作用。       方法        用改进的Feeney’s法制作大鼠脑冲击伤模型,分为正常组(A组)、颅脑创伤假手术组(B组)、颅脑创伤组(C组)、CICES治疗组(D组),观察打击后生存时间及术后6 h、1、3、7 d神经功能改变,分别检测血清IL-1β、TNF-α、脑组织补体C3水平及脑组织含水量。       结果        D组生存时间及神经功能评分(1、3 d)均优于C组(P<0.05),D组各检测时间点血清IL-1β、TNF-α浓度(6 h组IL-1β浓度除外)均低于C组(P<0.05,P<0.01),C3补体免疫组化染色结果显示:各检测时间点D组细胞膜外比C组有更少的补体附着(P<0.05),C组脑组织含水量(1、3、7 d)比D组更显著(P<0.05,P<0.01)。      结论        CICES能够显著减轻脑损伤后的炎性反应,在继发性脑损伤中起到重要的保护作用。
Abstract:
Objective       To determine the protective effects of the complement inhibiting component in Ephedra sinica(CICES) after traumatic brain injury (TBI) in rats.        Methods        Male SD rats were randomly divided into normal control, sham-operation group, TBI group, and CICES treatment group. Modified Feeney’s method was used to establish the rat model of experimental TBI with an impact of a weight of 30 g from 20 cm after craniotomy. CICES at 12 mg/kg was given intragastrically once per day for 3 d before the impact, while the rats of TBI group received same volume of normal saline. Survival time was observed and neural function at 6 h, and 1, 3 and 7 d after injury was assessed with Bederson grading system. ELISA was used to test the serum levels of  IL-1β and TNF-α. Immunohistochemical staining was used to detect the changes of complement 3 (C3) expression in the brain. The brain water content was measured.       Results        CICES treatment resulted in longer survival time and higher neurological functional scores than TBI group in 1 and 3 d after injury (P<0.05), and lower serum levels of IL-1 and TNF-α (in all time points except for 6 h for IL-1  level) compared with TBI group (P<0.05, 0.01). Immunohistochemical assay indicated that there were lesser C3 adhering on cell membrane in CICES treatment group than in TBI group(P<0.05). Brain water content (1, 3, and 7 d) was significantly higher in TBI group than in CICES treatment group (P<0.05, 0.01).       Conclusion        CICES significantly reduces the inflammatory reaction after brain injury, and plays an important protective role in secondary brain damage.
