摘要:
目的      研究胃转流术对2型糖尿病大鼠肝脏FoxO1表达的影响。      方法       40只SD大鼠随机分为糖尿病手术组(A组)、假手术组(B组)、正常对照组(C组)、糖尿病对照组(D组),每组10只。C、D组按A、B组术前和术后4、8、16、24周检测空腹血糖、空腹和餐后血清胰岛素、胰高血糖素样肽(GLP-1)。24周后处死各组大鼠,免疫组化和Western blot检测各组肝脏中FoxO1的表达。      结果       A组术后4周空腹血糖由术前的(20.2±2.1)mmol/L降至(10.5±2.5)mmol/L,术后24周空腹血糖为(7.7±1.1)mmol/L,术后4周空腹胰岛素由术前(2.87±0.08)mU/L增至(2.91±0.08)mU/L,术后24周空腹胰岛素为(3.00±0.06)mU/L;术后4周空腹血清GLP-1由术前(8.7±0.9)pmol/L升至(13.2±1.0)pmol/L,术后24周为(23.5±0.2)pmol/L。A组术后24周肝脏FoxO1表达量为18.4±1.2,B组为37.6±0.5,C、D两组分别为9.2±1.0、35.8±0.4,A组术后FoxO1的表达量明显低于B、D组,差异有统计学意义(P<0.05)。A组血清GLP-1浓度与肝脏FoxO1的表达量成负相关(r=-0.864,P<0.05)。      结论       胃转流术可显著降低2型糖尿病大鼠肝脏FoxO1的表达,降低血糖。
Abstract:
Objective       To investigate the effect of gastric bypass on FoxO1 expression in the liver of rats with type 2 diabetes, and to explore the mechanism for gastric bypass ameliorating type 2 diabetes.       Methods       Forty SD rats were randomly divided into a diabetes operation group (group A, n=10), a diabetes sham operation group (group B, n=10), a normal control group (group C, n=10) and a diabetes group (group D, n=10). The levels of fasting blood glucose (FBG), fasting and postprandial insulin and glucagon-like peptide-1(GLP-1) were measured before operation and at 4, 8, 16 and 24 weeks after operation. All the rats were sacrificed after 6 months, and the expression of FoxO1 in rat livers was determined by immunohistochemistry and Western blotting.       Results       As for the group A, the level of FBG decreased from 20.2±2.1 mmol/L before operation to 10.5±2.5 mmol/L at 4 weeks and 7.7±1.1 mmol/L at 24 weeks after operation, respectively. The level of fasting insulin increased from 2.87±0.08 mU/L before operation to 2.91±0.08 mU/L at 4 weeks and 3.0±0.06 mU/L at 24 weeks after operation, respectively. The level of fasting GLP-1 increased from 8.7±0.9 pmol/L before operation to 13.2±1.0 pmol/L at 4 weeks and 23.5±0.2 pmol/L at 24 weeks after operation, respectively. The expression level of FoxO1 was significantly lower in the group A (18.4±1.2) than in the group B (37.6±0.5) and group D (35.8±0.4) (P<0.05), and the FoxO1 expression was positively correlated with the serum level of GLP-1 in the group A (r=-0.864, P<0.05).       Conclusion       Gastric bypass can significantly reduce the levels of blood glucose and FoxO1 expression in the liver of rats with type 2 diabetes.
