摘要:
目的      观察荔枝核总黄酮(total flavone from Litchi chinensis Sonn,  TFL) 预防大鼠肝纤维化作用及活化肝星状细胞(hepatic stellate cells, HSC)凋亡的变化,从细胞凋亡角度研究二者之间的关系。      方法      以二甲基亚硝胺腹腔注射制作大鼠肝纤维化模型,同时以秋水仙碱为阳性对照,TFL[100、200 mg/(kg·d)]灌胃给药,6周后处死大鼠,放射免疫法(RIA)检测大鼠血清中透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)蛋白水平,取肝脏同一部位行HE、Masson染色观察大鼠肝纤维化程度,采用α-SMA及TUNEL免疫组化双重染色标记活化HSC凋亡。      结果      TFL高剂量组和秋水仙碱组血清肝纤维化指标(HA、LN、PCⅢ)均明显低于模型组 ( P<0.05)。与模型组比较,TFL高剂量组可明显增加活化的HSC凋亡指数,改善大鼠肝纤维化程度 (P<0.05)。      结论      荔枝核总黄酮可抑制肝纤维化大鼠HA、LN、PCⅢ表达,诱导HSC凋亡并改善二甲基亚硝胺所致肝纤维化程度,且具有一定量效关系。
Abstract:
Objective      To determine the prophylactic effect of total flavone from Litchi chinensis Sonn (TFL) on rats hepatic fibrosis and apoptosis of activated hepatic stellate cells (HSCs) in the livers, and to investigate the relationship between the prophylactic effects and activated HSC.       Methods      A rat model of hepatic fibrosis was established by intraperitoneal injection of dimethylnitrosamine. TFL was orally given to fibrotic rats at a dose of 100 or 200 mg/(kg·d). Colchicine was used as a positive control. After 6 weeks, all rats were sacrificed and their left liver lobes were examined histopathologically. The degrees of fibrosis were evaluated by HE and Masson straining. Serum trail levels of hyaluronic acid (HA), laminin (LN) and procollagen type Ⅲ (PCⅢ) were measured by radioimmunoassay(RIA); TUNEL-α-SMA methods was used to detect the apoptosis of HSC.       Results      Serum trail levels of HA, LN and PCⅢ were significantly lower in the high-dose TFL group and than in the colchicine group (all P<0.05). Degree of hepatic livers in the high-dose group were lesser severe in comparison with those of the control group. The apoptosis rates of HSC were also increased.       Conclusion      TEL inhibits the expression of HA, LN and PCⅢ, induces HSCs apoptosis and attenuates the degree of fibrosis in a dose dependent manner.
