摘要:
目的    观察益气滋肾合剂对牙周炎大鼠外周血TNF-α和IL-1β水平的影响,探讨其治疗牙周炎的机制。    方法      选用80只牙体牙列完整,无龋坏及牙周病的5月龄Wistar大鼠,雌雄各半,体质量200~250 g,随机分为5组:正常对照(n=10,N)组;模型对照(n=15,A)组,采用“牙颈部钢丝结扎+高糖饮食+激素”局部与全身因素相结合的方法,建立牙周炎动物模型;益气滋肾合剂(灌胃)高剂量[20 g/(kg·d),B1]、中剂量[10 g/(kg·d),B2]、低剂量[500 mg/(kg·d),B3]组(n=15)。12周后通过对大鼠一般状态、牙齿松动度、大鼠外周血中IL-1β、TNF-α含量的变化和大鼠牙槽骨微观形态分析,评价不同浓度的益气滋肾合剂治疗大鼠牙周炎的效果。    结果      大鼠外周血中TNF-α、IL-1βA组[(4.909±0.505),(0.318±0.086)]显著高于B1[(4.051±0.463),(0.253±0.193)]、B2[(4.085±0.374),(0.266±0.062)]组(P<0.05);Micro-CT成像分析得出骨小梁数量和骨小梁分离度值A组[(1.24±0.22),(789.3±157.0)]显著低于B1[(1.95±0.34),(362.3±62.7)]、B2[(1.88±0.39),(396.2±58.2)]组(P<0.05)。    结论      益气滋肾合剂可以降低大鼠外周血中IL-1β、TNF-α的水平,增加牙槽骨密度,对大鼠牙周炎具有治疗作用。
Abstract:
Objectives      To determine the effect of Tonifying Qi Nourishing Kidney Mixture on the level of tumor necrosis factor-α(TNF-α)and cytokine interleukin-1β(IL-1β) in the peripheral blood and on morphological changes of alveolar bone during experimental periodontitis.     Methods      Eighty 5-months-old Wistar rats were divided into 5 groups at random. The rats of Group N was killed at the beginning of the experiment and used as the normal control. And the animals of Group A, B1, B2 and B3 were inflicted to be periodontitis models by ligating dental cervix with steel wire, high-glucose feeding and topical hormone treatemnt. The rats in group A were killed at the end of periodontitis establishment to assess the features of the model. The rats fo Group B1, B2 and B3 received an intragastrical injection of the mixture at 20, 10 and 5 g/(kg·d) respectively for 12 weeks and then were killed after general evaluation and blood sampling. The levels of TNF-α and IL-1β in the peripheral blood were measured with enzyme-linked immunosorbent assay. Morphological changes of alveolar bone was observed by microcomputed tomography (micro-CT).     Results      The levels of TNF-α and IL-1β in treated group B1 (4.909±0.505 and 0.318±0.086) and group B2(4.085±0.374 and 0.266±0.062)were significantly lower than those in the control group A (4.909±0.505 and 0.318±0.086, P<0.01). At the same time, trabecular number and trabecular separateion in the experimental group B1 (1.95±0.34 and 362.3±62.7)and group B2(1.88±0.39 and 396.2±58.2)were significantly better than those in the control group A (1.24±0.22 and 789.3±157.0, P<0.01).     Conclusion      Tonifying Qi Nourishing Kidney Mixture reduces the levels of TNF-α and IL-1β in the peripheral blood, and enhances the density of alveolar bone, thus exerting therapeutic effect on periodontitis.
