目的    研究Rho亚家族蛋白(RhoA、B、C)与Rho激酶即ROCK(ROCKⅠ、Ⅱ)在大鼠不同孕期胎盘中的表达水平及其动态变化特点。    方法     正常受孕的20只雌性大鼠分为孕7、10、13、15、18 d组;各组孕鼠正常饲养,并分别于妊娠第7、10、13、15、18天剖宫取胎盘,测量胎盘湿重及胎盘直径,并记录胎盘个数;采用Western blot及免疫组化法测定胎盘组织中RhoA、B、C与ROCKⅠ、Ⅱ蛋白的表达水平。    结果     随着孕龄增加,大鼠胎盘的质量和直径都在增加;Western blot及免疫组化法检测结果显示,RhoA、RhoC的表达水平在孕13、15、18 d组显著高于孕7、10 d组(P<0.01),而RhoB表达水平无明显差异(P>0.05)。ROCKⅠ、ROCKⅡ在孕18 d组表达显著高于孕7、10、13、15 d组(P<0.01)。    结论     RhoA、RhoC、ROCKⅠ、ROCKⅡ在大鼠胎盘组织中的表达呈上升趋势,而RhoB则无明显变化;Rho/ROCK信号通路在正常妊娠不同时期的大鼠胎盘组织中发挥重要作用。
Abstract:
Objective      To determine the expressions and the dynamic changes of Rho proteins (RhoA, RhoB and RhoC) and their kinase ROCK (ROCKⅠ and ROCKⅡ) in rat placenta at different stages of gestation.     Methods      A total of 20 pregnant female rats were randomly divided into 5 groups (n=4). Their placents were obtained when the rats were pregnant at 7, 10, 13, 15, and 18 d respectively. Wet weight and diameter of the placenta were measured, and the number of the placenta was counted and recorded. The expression levels of RhoA, RhoB, RhoC, ROCKⅠ and ROCKⅡ were detected by immunohistochemical staining and Western blot analysis.     Results      With the increase of gestational period, the weight and diameter of the placenta were increased. The expression levels of RhoA and RhoC in 13, 15, and 18 d of pregnant groups were significantly higher than those in 7 and 10 d of pregnant groups (P<0.01), but the expression of RhoB had no obvious difference among these groups (P>0.05). ROCKⅠ and ROCKⅡ in 18 d of pregnant group was expressed significantly higher than those in 7, 10, 13 and 15 d of pregnant groups (P<0.01).     Conclusion     The expression levels of RhoA, RhoC, ROCKⅠ and ROCKⅡ are increased in rat placental tissues with the elapse of pregnant period, but that of RhoB has no such significant change. These results indicate that Rho/ROCK signaling pathway may play an essential role in the development of placenta during normal pregnancy.
