摘要:
目的    探讨核干因子(nucleostemin, NS)和增殖细胞核抗原(proliferating cell nuclear antigen, PCNA)在结直肠癌组织中的表达与血清叶酸水平的关系。    方法    2009年3-12月在肠镜下收集50例结直肠癌患者的组织,根据国际结直肠癌的病理分类标准分为高分化组(12例)、中分化组(17例)、低分化组(21例)和正常肠黏膜组(50例),同时取受检者静脉血5 ml;采用免疫组织化学法检测NS和PCNA结直肠癌组织和正常肠黏膜的表达,采用磁性颗粒化学发光免疫法检测血清叶酸水平。    结果    NS和PCNA在结直肠癌阳性表达率分别是70.0%(35/50)和72.0%(36/50),两者在正常肠黏膜中未见表达。NS和PCNA在高分化结直肠癌阳性率为41.7%(5/12)和33.3%(4/12),分别显著低于中分化结直肠癌阳性率[76.5%(13/17)和82.4%(14/17)]与低分化结直肠癌阳性率[81.0%(17/21)和85.7%(18/21),P<0.05]。患者血清叶酸水平在结直肠癌NS和PCNA阳性组与阴性组无显著差别 (P>0.05)。    结论    NS和PCNA在结直肠癌组织中有表达,且与结直肠癌的恶性程度密切相关,但血清叶酸水平与NS和PCNA在结直肠癌组织中的表达无关。
Abstract:
Objective    To investigate the correlation between the expression of nucleostemin (NS) and proliferating cell nuclear antigen (PCNA) in colorectal cancer tissues and serum folic acid concentrations.     Methods    From March to December 2009, colorectal cancer tissues were collected from 50 patients through colonoscopy. According to international colorectal cancer classification standard for the pathological, which divided into well-differentiated group(n=12), moderately differentiated (n=17), poorly differentiated(n=21)and corresponding normal intestinal mucosa tissues. At the same time, venous blood of 5 ml was collected from each of the above patients. Immunohistochemistry was used to detect the expression of NS and PCNA in colorectal cancer tissues and normal intestinal mucosa tissues. Serum folic acid concentrations were determined with magnetic chemiluminescence immunoassay.     Results    The positive expression rates of NS and PCNA in colorectal cancer tissues were 70% (35/50) and 72% (36/50), while neither of NS and PCNA expression was detected in the normal intestinal mucosa tissues. The positive expression rates of NS and PCNA in well differentiated colorectal cancer tissues were 41.7% (5/12) and 33.3% (4/12), which were significantly lower than those [76.5%(13/17) and 82.4%(14/17)] in the  moderately differentiated colorectal cancer tissues and [81.0% (17/21) and 85.7% (18/21)] in the poorly differentiated colorectal cancer tissues (P<0.05). There was no significant difference in serum folic acid concentration between NS and PCNA positive expression group and negative expression group (P>0.05).     Conclusion    NS and PCNA are expressed in colorectal cancer tissues, and their expression levels are correlated with the malignant degree of colorectal cancer. Serum folic acid concentration is not related to NS and PCNA expression in colorectal cancer.
