摘要:
目的    观察生存素(survivin)反义寡核苷酸(anti-survivin oligonucleotides, ASODN)对人胃癌细胞株BGC-823增殖、凋亡、侵袭、迁移能力的影响。    方法      脂质体介导ASODN或对照SODN转染人胃癌细胞株BGC-823后,RT-PCR方法检测survivin的mRNA表达水平,四氮唑盐(MTT)法检测ASODN或SODN对肿瘤细胞的增殖抑制效果,Transwell小室法检测细胞侵袭和迁移能力的改变,流式细胞术检测细胞周期及凋亡指数的改变。     结果       ASODN转染可明显降低survivin 的表达(P<0.05),并且ASODN转染具有以浓度及时间依赖的方式抑制BGC-823细胞增殖的作用(P<0.05)。用600 nmol/L浓度的ASODN转染BGC-823后,BGC-823细胞迁移和侵袭能力下降(P<0.05),细胞凋亡指数(apoptotic index,AI)由(1.62±0.36)增加到(11.31±0.38)(P<0.01),而增殖指数(proliferation index,PI)则由(43.80±0.81)下降至(27.77±2.97)(P<0.05)。停滞于G2/M期的细胞明显增加(P<0.01)。    结论      下调survivin表达的ASODN可通过抑制胃癌细胞增殖、诱导凋亡、降低胃癌细胞侵袭和迁移能力而发挥抗肿瘤作用。
Abstract:
Objective    To determine the effects of anti-survivin oligonucleotides (ASODN) on the proliferation, apoptosis, invasion and migration in gastric cancer BGC-823 cells.     Methods      After ASODN or SODN was introduced by LipofectamineTM 2000 transfection reagent into BGC-823 cells, the mRNA expression of survivin, was detected by RT-PCR, cell proliferation was measured by 4-methyl-tetrazolium (MTT) assay, cell invasion and migration were assayed by Transwell chamber tests, and cell cycle and apoptosis index (AI) was detected by flow cytometry.     Results      ASODN transfection resulted in a marked decrease of survivin expression (P<0.05). And the cell proliferation was inhibited by ASODN transfection in a dose- and time-dependent manner (both P<0.05). Both cell invasion and migration activities of BGC-823 were decreased (P<0.05) after the transfection of 600 nmol/L ASODN. AI of ASODN transfection group was increased significantly when compared with that of control group (11.31±0.38 vs 1.62±0.36, P<0.01), and proliferation Index (PI) was decreased significantly in ASODN transfection group than in control group (27.77±2.97 vs 43.80±0.81, P<0.05). Cells in G2/M stage were increased in ASODN transfection group compared with control group (P<0.01).     Conclusion     Down-regulation of survivin by ASODN inhibits the invasion and migration of BGC-823 cells through suppressing cell proliferation and improving cell apoptosis.
