摘要:
目的   探讨MRL/lpr小鼠中CD4+CD25+调节性T细胞(Treg细胞)的含量、增殖能力及其对效应性T淋巴细胞(Teff细胞)的抑制能力及意义。   方法   流式细胞检测法检测8只MRL/lpr小鼠及8只BALB/c小鼠脾脏中Treg细胞的水平;磁珠分选法分选出脾脏中的Treg细胞与Teff细胞,离体培养2周后,分别测量Treg细胞数量,并比较差异;用Treg细胞与Teff细胞组成共培养体系,5 d后检测Teff细胞的水平,计算Treg细胞对Teff细胞的抑制率并比较差异。   结果   MRL/lpr小鼠脾脏中Treg细胞的总含量与BALB/c小鼠无统计学差异[(4.88±0.55)% vs(5.03±0.34)%,P>0.05];离体培养2周后MRL/lpr小鼠的Treg细胞数量显著低于BALB/c小鼠[(6.61±0.92)×105 vs (13.25±1.91)×105P<0.05];共培养5 d后MRL/lpr小鼠Treg细胞对Teff细胞的抑制率显著低于BALB/c小鼠Treg细胞(P<0.05)。   结论   MRL/lpr小鼠体内Treg细胞增殖能力、抑制能力的下降可能导致体内免疫平衡的破坏、自身抗原-抗体反应,参与MRL/lpr小鼠的系统性红斑狼疮的发病。
Abstract:
Objective   To investigate the percentage, reproductive ability of CD4+CD25+ regulatory T cells (Tregs) in MRL/lpr mice, and their suppressive activity to effective T cells (Teffs).   Methods   Flow cytometry was used to determine cell populations of Tregs in the spleen of 8 MRL/lpr mice and 8 BALB/c mice respectively. Magnetic beads were employed to separate Tregs and Teffs from the splenic cells of the 2 kinds of mice. Then the Tregs were cultured in vitro for 2 weeks and cell populations were determined. The Tregs were cultured with Teffs for 5 d and then the level of Teffs was detected. The suppresive rate of Tregs to Teffs was calculated.   Results   No significant difference was detected in cell populations of Tregs between 2 kinds of mice [(4.88±0.55)% and (5.03±0.34)%, respectively; P>0.05]. After cultivation in vitro for 2 weeks, cell populations of Tregs were significantly lower in MRL/lpr mice than that in BALB/c mice [(6.61±0.92)×105 and (13.25±1.91)×105, respectively; P<0.05]. After co-cultivation in vitro for 5 d, the suppressive rate of Tregs to Teffs was significantly lower in MRL/lpr mice than that in BALB/c mice (P<0.05).   Conclusion The Tregs in MRL/lpr mice have decreased reproductive ability and suppressive activity, which may lead to impairment of immune balance and induction of auto-antibody, and thus the illness of systemic lupus erythematosus .
