目的   分析丝氨酸-苏氨酸激酶受体相关蛋白在系统性红斑狼疮(systemic lupus erythematosus, SLE)患者外周血单个核细胞中的表达水平,探讨该蛋白检测在系统性红斑狼疮病情判断中的可能应用。   方法   实验分为3组:SLE稳定组、SLE活动组和健康对照组,每组6例,分别收集其外周血单个核细胞,用相对和绝对定量的等量异位标签串联质谱技术对丝氨酸-苏氨酸激酶受体相关蛋白进行鉴定和相对定量分析,用Western blot技术对该蛋白的表达水平进行较大样本的独立验证。   结果   基于质谱分析的蛋白组学结果表明,丝氨酸-苏氨酸激酶受体相关蛋白在系统性红斑狼疮活动期患者外周血单个核细胞中表达显著下调(与正常人的相对比值为0.290 6),且该蛋白的低表达为Western blot实验进一步证实。临床资料分析表明,该蛋白的表达水平与系统性红斑狼疮疾病活动指数呈负相关(r=-0.607,P<0.05)。   结论   丝氨酸-苏氨酸激酶受体相关蛋白可能参与SLE的发病,该蛋白有望作为系统性红斑狼疮的活动阶段特异性标志物。
Abstract:
Objective    To analyze the expression level of serine-threonine kinase receptor-associated protein (STRAP) in peripheral blood mononuclear cells of patients with systemic lupus erythematosus (SLE) and to investigate the possible application of STRAP test in evaluation of SLE.    Methods     Peripheral blood mononuclear cells were collected from 6 stable and 6 active SLE patients and 6 healthy controls, respectively. STRAP was first identified and quantified with isobaric tagging for relative and absolute protein quantification (iTRAQ) in combination with tandem mass spectrometry, and then confirmed by Western blotting in a larger independent sample size.    Results    Initial mass spectrometry-based proteomic analysis revealed that the STRAP level was significantly lower in active SLE patients than in healthy controls with a relative ratio of 0.290 6, which was further confirmed by Western blotting (P<0.05). Clinical data analysis disclosed that the expression level of STRAP in active SLE patients was negatively correlated with the SLE activity index (r=-0.607, P<0.05).    Conclusion     STRAP may be involved in the pathogenic process of SLE and can be used as a potential condition-specific indicator of active SLE.
