摘要:
目的    研究GSK3特异性抑制剂氯化锂(lithium chloride,LiCl)对尾吊大鼠骨密度的影响。    方法    SD雄性大鼠18只,分为3组:正常对照组(Control)、尾吊对照组、尾吊LiCl组,每组6只。尾吊LiCl组大鼠每日灌胃给予LiCl 20 mg/(kg·d)。分别于第1天和第15天使用双能X线骨密度测量仪测定大鼠左股骨骨密度(bone mineral density,BMD);第15天处死大鼠取左胫骨近心端进行组织包埋切片,HE染色,进行骨组织计量学测量,包括骨小梁面积百分率 (Tb.Ar%)、骨小梁数量 (Tb.N)、骨小梁平均厚度(Tb.Th);对大鼠右股骨进行三点弯曲实验,测量骨力学指标。采用Western blot观察骨组织中β-catenin蛋白的表达。    结果    实验结束后,尾吊对照组和尾吊LiCl组大鼠BMD均低于正常对照组(P<0.05),而尾吊对照组大鼠BMD明显低于尾吊LiCl组(P<0.01);骨组织计量学结果,尾吊对照组大鼠骨小梁平均厚度低于正常对照组(P<0.05),骨小梁面积百分率、骨小梁数量明显低于正常对照组(P<0.01),而尾吊LiCl组大鼠三项参数均高于尾吊对照组(P<0.05);骨力学测试结果,尾吊对照组大鼠股骨弹性模量低于正常对照组(P<0.05),刚性系数明显低于正常对照组(P<0.01),而尾吊LiCl组大鼠股骨弹性模量高于尾吊对照组(P<0.05),刚性系数明显高于尾吊对照组(P<0.01)。Western blot结果显示,尾吊对照组β-catenin蛋白表达量明显低于正常对照组,而尾吊LiCl组β-catenin 蛋白表达量明显高于尾吊对照组。    结论    尾部悬吊能够造成大鼠废用性骨丢失,而LiCl能有效地预防尾吊所导致的骨质疏松。
Abstract:
Objective    To investigate the effect of lithium chloride (LiCl), a special inhibitor of GSK-3 in Wnt signal pathway, on the bone loss of hindlimb unloading rats.     Methods    Eighteen male SD rats were randomly divided into 3 groups: control group, hindlimb unloading (HU) group, and HU+LiCl group. The HU+LiCl group were intragastric administrated with LiCl [20 mg/(kg·d)]. Bone density (BMD) of the rats’ left femurs on the 1st and 15th days was measured by dual energy X-ray absorptiometry (DEXA). On day 15, all rats were sacrificed to remove the proximal part of the left femur for HE staining. Then the morphological changes were observed, and bone histomorphometry including percent trabecular area (Tb.Ar%), trabecular number (Tb.N), and trabecular thick (Tb.Th) was carried out. Rats’ right femurs were resected to measure the bone biomechanical indexes in threepoint bending experiments. The expression of β-catenin protein of bone was tested by Western blotting.     Results    BMD of the HU group and HU+LiCl group were lower than that of the control group (P<0.05), while that of the HU group were significantly lower than the HU+LiCl group (P<0.01). Tb.Th of HU group was lower than that of control group (P<0.05), while the Tb.Ar% and Tb.N of HU group were significantly lower than those of control group (P<0.01), but these 3 indexes of the HU+LiCl group were higher than HU group (P<0.05). The elastic modulus of HU group were lower than control group (P<0.05), and the stiffness coefficient of HU group were significantly lower than control group (P<0.01), while the elastic modulus of HU+LiCl group were higher than those of HU group  (P<0.05) and the stiffness coefficient of HU+LiCl group were significantly higher than HU group  (P<0.01). Western blotting showed that the expression of β-catenin of HU group was lower than control group, while HU+LiCl group was higher than HU group.     Conclusion    Hindlimb unloading can cause bone loss in rats with disuse. LiCl may be used to prevent rats’ osteoporosis caused by hindlimb unloading.
