摘要:
目的 研究RNA解旋酶p68在真皮多能干细胞(dermal multipotent stem cells, DMSCs)增殖过程中的表达特点,并探讨其在创伤愈合中的意义。方法 通过免疫细胞化学和Western blot方法检测p68在DMSCs不同生长状态的表达特点,早期伤口液刺激对DMSCs中p68表达的影响并通过MTT法检测DMSCs增殖的变化,进一步通过RNAi抑制p68表达之后检测细胞增殖的变化特点。 结果 增殖活跃的DMSCs中p68表达较强,饥饿以后,p68表达降低;伤后1 d伤口液可明显促进DMSCs中p68的表达及细胞增殖(P<0.01);RNAi抑制p68表达之后DMSCs增殖明显受抑(P<0.01)。 结论 RNA解旋酶p68参与DMSCs的增殖调控,创伤微环境在修复细胞的启动中可能发挥作用。
Abstract:
Objective To study the function of RNA helicase p68 in the proliferation of dermal multipotent stem cells (DMSCs).  Methods The expression of p68 protein in DMSCs at different growth states and pre-treated with wound fluid was detected by immunocytochemical assay and Western blot analysis. After the down-regulation of p68 expression by sequence specific siRNA, the proliferation of DMSCs was evaluated by MTT assay.  Results p68 expression was higher in DMSCs of exponential growth than that of cells in contact inhibition. The wound fluid induced the p68 expression and proliferation of DMSCs. When p68 was down-regulated by siRNA, the proliferation of DMSCs was significantly inhibited.  Conclusion The expression of p68 in DMSCs is involved in the regulation of DMSCs proliferation, and the microenvironment of wound may play a crucial role in the initiation of proliferation of wound repair cells.
