摘要:
目的   通过研究乌司他丁对TNF-α作用的A549细胞ENaC-β转录和翻译水平的影响。   方法   将A549细胞分组为空白组、乌司他丁组、TNF-α组和乌司他丁+TNF-α组,TNF-α浓度为100 ng/ml,各组作用时间均为6、12、24 h。乌司他丁组及乌司他丁+TNF-α组在12 h时间点上分别用1 250、2 500、5 000 U/ml 3种浓度乌司他丁作用A549细胞,采用浓度5 000 U/ml乌司他丁分别作用6、24 h,RT-PCR半定量检测ENaC-β mRNA表达量,Western blot半定量检测ENaC-β蛋白浓度。   结果   ①在作用不同时间后,乌司他丁对ENaC-β mRNA和蛋白表达水平的影响与空白组比较差异无统计学意义(P>0.05)。②5 000 U/ml乌司他丁+TNF-α组分别作用6、12、24 h后ENaC-β mRNA及蛋白表达较TNF-α组均明显升高(P<0.01)。③作用12 h后,各浓度乌司他丁+ TNF-α组ENaC-β mRNA及蛋白表达较TNF-α组均明显升高(P<0.01),且各浓度组间ENaC-β mRNA及蛋白表达差异也有统计学意义(P<0.05)。   结论   乌司他丁不能直接上调A549细胞ENaC-β转录和翻译水平,但能以浓度依赖方式对抗炎症反应对ENaC-β表达的抑制作用。
Abstract:
Objective   To study the effect of ulinastatin on transcript and translation of ENaC-β mRNA and protein of human alveolar epithelial A549 cells treated with TNF-α and its mechanism of action in treatment of acute respiratory distress syndrome.    Methods   A549 cells were divided into normal control group, ulinastatin group, TNF-α group and ulinastatin + TNF-α group. TNF-α concentration was 100 ng/ml. A549 cells in each group were treated with ulinastatin,TNF-α or ulinastatin + TNF-α for 6, 12 and 24 h, respectively. A549 cells in ulinastatin and ulinastatin + TNF-α groups were treated with ulinastatin at the concentration of 1 250, 2 500 and 5 000 U/ml for 12 h, and then at the concentration of 5 000 U/ml for 6 and 24 h, respectively. ENaC-β mRNA and protein expression levels were measured by semi-quantitative RT-PCR and Western blotting.    Results   No significant difference was found in effect of ulinastatin on ENaC-β mRNA and protein expression level in A549 cells in 6, 12 and 24 h after treatment with it between treatment group and control group(P>0.05). The ENaC-β mRNA and protein expression level in A549 cells in 6, 12 and 24 h after treatment with ulinastatin (at the concentration of 5 000 U/ml) + TNF-α was significantly increased compared with that after treatment with TNF-α (P<0.01). However, it was significantly different 12 h after treatment with different concentrations of ulinastatin + TNF-α from that after treatment with TNF-α alone(P<0.01). The ENaC-β mRNA and protein expression level was different at different time points after treatment with different concentrations of ulinastatin + TNF-α(P<0.05).    Conclusion   Ulinastatin can not directly up-regulate the transcription and translation of ENaC-β in A549 cells, but can reverse the inhibitory effect of inflammatory response on ENaC-β expression in a concentration-dependent manner.
