摘要:
目的    建立db/db小鼠早期糖尿病肾病(diabetic nephropathy,DN)微小RNA(microRNA,miRNA)的差异表达谱。    方法    采用12只9周龄高血糖且24 h尿白蛋白显著增高的db/db小鼠作为早期糖尿病肾病组,12只9周龄db/m小鼠作为正常对照组,用于miRNA芯片和实时荧光定量RT-PCR检测。Trizol法抽提肾脏总RNA,YM-100微离心滤器富集小RNA,芯片技术检测DN组和对照组miRNAs的差异表达谱。应用实时荧光定量RT-PCR验证部分差异miRNAs,运用生物信息学技术预测部分相关miRNAs的靶基因。     结果    芯片结果显示,66个miRNA在DN组和对照组存在差异表达(P<0.01),其中35个miRNAs在DN组呈高表达,31个miRNAs在DN组呈低表达,差异倍数从1.08~10.20。实时荧光定量RT-PCR进一步验证部分miRNA的表达,结果显示3个miRNAs在DN组表达上调,分别为:miR-196a、miR-98和miR-29c;4个表达在DN组下调,分别为miR-21、miR-451、miR-709和miR-187,差异显著(P<0.01)。    结论    部分miRNAs在DN和正常对照肾脏组织中存在差异表达,可能参与了DN发生的分子机制。
Abstract:
Objective    To establish the differential expression profile of early diabetic nephropathy (DN)- related microRNA (miRNA) in kidneys of db/db mice.     Methods    Twelve 9-week-old male db/db mice with hyperglycemia and significantly elevated urinary albumin excretion were used as an early DN group,and another twelve 9-week-old male db/m mice were used as a control group. Total mRNA was extracted from kidney tissues of mice and miRNA was concentrated with the YM-100 micro-centrifugation filter. Differential expression profile of miRNA in DN and control groups was detected by miRNA assay and RT-PCR, respectively. Target gene of related miRNA was detected with bioinformatics technology.     Results    miRNA assay showed that 66 miRNAs were differentially expressed in DN and control groups (P<0.01). Of the 66 miRNAs, 35 and 31 were highly and lowly expressed, respectively, in DN group, with a difference of 1.08 to 10.20 folds. RT-PCR showed that the expression of 3 miRNAs was up-regulated (miR-196a, miR-98 and miR-29c) and the expression of 4 miRNAs was down-regulated (miR-21, miR-451, miR-709 and miR-187) in DN group(P<0.01).     Conclusion    Some miRNAs are differentially expressed in the kidney tissues of DN and normal mice, suggesting that miRNAs may be involved in the molecular mechanism of DN.
