摘要:
目的 探讨胸腺肽(thymosinαl,Tα1)对促进人脐静脉血管内皮细胞(human umbilical veinvessel endothelial cells, HUVECs)增殖的影响以及刺激血管内皮细胞生长因子(vascular endothelial growth factor, VEGF)的合成情况。 方法 体外培养HUVECs细胞系,采用四甲基偶氮唑蓝法(MTT)法测定HUVECs的增殖情况,以流式细胞仪检测细胞周期,Western blot检测VEGF在HUVECs 的合成。 结果 浓度为2.5、5、10 μg/ml的Tα1作用于HUVECs后,其对应的D(492)值、增殖率分别为(0.498±0.027)、(96.2±4.5)%,(0.737±0.018)、(182.0±5.8)%,(0.745±0.021)、(185.0±6.2)%与对照组比较,明显增高(P<0.01)。S期细胞增多。Western blot检测结果显示实验组VEGF条带灰度值/内参灰度值(0.29±0.01)与对照组(0.12±0.06)相比,显著上升(P<0.05)。 结论 Tα1能促进HUVECs的增殖及增强血管内皮细胞内VEGF的合成和分泌,在创伤组织修复过程中可能发挥促新生血管形成的作用。
Abstract:
Objective To investigate the effects of thymosin α1 on the proliferation of human umbilical veinssel endothelial cells (HUVECs) and synthesis of vascular endothelial growth factor (VEGF).  Methods HUVECs (ECV-304) were cultured with the treatment of 100 μl thymosin α1 at 0, 5 or 2.5 μg/ml for 1 d in vitro. The proliferation of HUVECs were measured with MTT assay, cell cycle was tested with flow cytometry and the synthesis of VEGF was detected with Western blot analysis.  Results Thymosinal promoted the proliferation of HUVECs, and the cells in S-phase was increased, and obviously promoted the synthesis of VEGF.  Conclusion Thymosin α1 promotes the proliferation of HUVECs and increases the synthesis of VEGF in HUVECs, and may have the effect of increasing the angiogenesis in the process of wound tissue.
