摘要:
目的    观察1,25-(OH)2D3及复方丹参对异基因骨髓移植(Allo-BMT)后大鼠T细胞亚群和细胞因子的影响。    方法    以雄性SD大鼠为供鼠,雌性Wistar大鼠为受鼠,40只受鼠随机分成aGVHD组及干预组,干预组包括1,25-(OH)2D3组、复方丹参组、两药联合组,每组10只。观察各组T细胞亚群(CD4+、CD8+)和细胞因子(IL-2、IFN-γ、IL-4、IL-10)的变化。    结果    当aGVHD表现较明显时,CD4+、CD8+增高,以CD4+增高为主,与移植前比差异有统计学意义(P<0.05);经干预方案处理后CD4+ 、CD8+增高的幅度较aGVHD组低,差异有统计学意义(P<0.05),以CD4+更为明显;动态监测显示与Thl相关的细胞因子IL-2、IFN-γ在各干预组呈下降趋势,与Th2相关的细胞因子IL-10呈上升趋势,均在第21天变化较为明显,与第0天相比有统计学差异(P<0.05);IL-4无显著性变化。    结论    1,25-(OH)2D3及复方丹参能够降低CD4+T淋巴细胞的数量,具有调节Thl/Th2平衡,抑制Thl的增殖及功能发挥,促进Th2细胞增殖,发挥免疫耐受的作用。
Abstract:
Objective    To investigate the effect of 1,25-(OH)2D3and salvia miltiorrhiza on T cell subsets and cytokines in rats after allogeneic bone marrow transplantation.     Methods    Female Wistar rats were used as recipients and male SD rats were used as donors. All Wistar rats were divided into aGVHD group and intervention groups at random, and the intervention groups were further divided into 1,25-(OH)2D3 group, salvia miltiorrhiza group and combination group. The changes of T cell subsets (CD4+ and CD8+) and cytokine (IL-2, IFN-γ, IL-4 and IL-10) in every group were detected and measured.     Results    When the presentation of aGVHD was relatively conspicuous, CD4+ and CD8+were increased, and the increase of CD4+ was predominant. Compared with the difference prior to transplantation, the difference was statistically significant (P<0.05). After being treated with the intervention program, the increase extent of CD4+ and CD8+ was lower than that of aGVHD group (P<0.05), in which the increase of CD4+ was more conspicuous. Dynamic monitoring shows that the cytokine IL-2 and IFN-γ which were relevant to Th1 tended to go down in every intervention group, while the cytokine IL-10 relevant to Th2 tended to go up. The change was both relatively conspicuous on Day 21. Compared with the levels on Day 0, the difference was statistically significant (P<0.05). But there was no obvious change for IL-4.     Conclusion    1,25-(OH)2D3 and salvia miltiorrhiza can reduce the quantity of CD4+ T lymphocytes, regulate the balance of Thl/Th2, restrict the proliferation and function exertion of Thl, promote the proliferation of Th2 cells and exert immune tolerance.
