摘要:
目的   比较分析FGFR2 Ser252Trp突变小鼠(mutant,Mu)下颌骨和同窝对照野生型小鼠(wild type,WT)下颌骨的形状和发育差异性。   方法   采用三维坐标测量仪检测下颌骨11个标记点的三维坐标值,用欧几里德几何距离矩阵法(euclidean distance matrix analysis,EDMA)对突变小鼠和对照小鼠下颌骨进行形状和发育分析。   结果   4周时FGFR2 Ser252Trp突变小鼠下颌骨形状与同窝对照组相比整体有所缩小,但无统计学差异(P>0.05);8周时突变小鼠下颌骨形状缩小具有统计学差异(P<0.05),主要表现在冠状突和下颌角缩小并沿前后方向向前缩进,随着年龄的增加下颌角的缩进更明显。   结论   FGFR2 Ser252Trp点突变对小鼠下颌骨形状产生缩短畸形的影响,随着年龄的增加缩短影响有所加重。
Abstract:
Objective   To compare and analyze the morphologic and growth difference between the mandibles of the mouse carrying Ser252Trp mutation in fibroblast growth factor receptor (FGFR2) which was regarded as a model for human Apert syndrome, and of their wild type controls.    Methods   The 3-D coordinate data of 11 landmarks of the mandible were collected by a 3-D coordinate measuring machine. Data were analyzed using Euclidean distance matrix analysis (EDMA) to quantitatively compare the morphologic and growth difference between the mandibles of mutants and their controls.    Results   At 4 weeks old, the mutants’ mandible was smaller than their wild type control, and there was no statistical difference between them (P>0.05). The mandible was much smaller in the mutants than their wild type controls, with a statistical difference when they were 8 weeks old (P<0.05). The major abnormality in the mutants was the anteriorly displaced coronoid process and mandibular angle along anterio-posterior axis, which becomes more severe with age.    Conclusion   The reducing of the mutants’ mandibles becomes more severe with age.
