摘要:
目的    观察中药制剂养精种子汤对缺氧雄性大鼠生精细胞凋亡的影响。  方法   36只雄性SD大鼠分为正常组、缺氧组、养精种子汤低剂量(10 g·kg-1·d-1)组和养精种子汤高剂量(35 g·kg-1·d-1)组(n=9)。用模拟海拔5 000 m的低压舱,造成大鼠生殖系统损伤模型,应用光镜、电镜、原位末端标记法(TUNEL)观察服用养精种子汤前后大鼠生精细胞凋亡情况。    结果    光镜下缺氧组示生精小管内生殖细胞排列紊乱,生精细胞数减少,生精上皮变薄,管腔内精子数明显减少,而养精种子汤高剂量组可见生精小管各级生精细胞发育正常,排列整齐,管腔内有大量成熟精子。电镜下缺氧组出现明显细胞凋亡情况,养精种子汤高剂量组明显改善。TUNEL法检测表明缺氧组与正常组、养精种子汤低剂量组和养精种子汤高剂量组相比生精细胞凋亡率明显增高(P<0.01)。    结论    养精种子汤能明显减少缺氧雄性大鼠的生精细胞凋亡。
Abstract:
Objective    To study the effects of Yangjing Zhongzi decoction (YJZZ) on the apoptosis of spermatogenic cells in male rats after hypoxic injury.     Methods    The total of 36 Sprague-Dawley(SD) male rats were randomly divided into normal group, hypoxic group, high-dose YJZZ group and low-dose YJZZ group. The rats of the later 3 groups lived for 4 weeks in a hypobaric chamber simulating 5 000 m altitude. Besides fed with standard chow, the rats of high-dose and low-dose YJZZ groups were given the decoction intragastrically at a dose of 35 and 10 g·kg-1·d-1 respectively for 4 weeks. Then the rats were sacrificed and their testis were resected for light microscopic and electron microscopic studies. The apoptosis of spermatogenic cells were detected with terminal deoxynucleotidyl transferase-mediated dUDP-nick end labeling (TUNEL) method.     Results    Compared with normal control, the testes in the hypoxic group had germ cells in a disordered arrangement, decreased spermatogenic cells and sperms in the seminiferous tubules, and thinned seminiferous epithelium. While the large-dose YJZZ group did not show such morphological features. In the hypoxic group, electron microscopy observed obvious apoptosis in spermatogenic cells, but no apoptosis was seen in the large-dose YJZZ group. The percentage of positive TUNEL cells in the hypoxic group was significantly higher than those in the other groups (P<0.01).     Conclusion    YJZZ decoction can reduce the apoptosis in spermatogenic cells and increase the number of spermatogenic cells.
