摘要:
目的    采用64层MSCT灌注成像(CTP)观测大鼠C6脑胶质瘤灌注参数的动态变化规律,探讨大鼠C6脑胶质瘤的血管生成特征。    方法    采用立体定向仪进行C6细胞脑内接种,建立大鼠C6脑胶质瘤模型。每次随机抽取10只接种鼠分别对应于5~9 d、10~14 d、15~19 d 3个时间段行CTP及病理学检查,观测大鼠C6脑胶质瘤血流灌注参数的变化规律。    结果    大鼠脑内C6细胞接种后5~9 d,瘤内CBF和CBV值即有增高,并随时间而继续上升,于10~14 d达到高峰,并稳定在一较高水平,15~19 d肿瘤CBF和CBV值有所下降。肿瘤PS值在5~9 d内即有明显增高,10 d以后继续增高,15 d以后增高显著,并在20 d内无下降趋势。而肿瘤组织的MTT不随时间的延长而发生变化。    结论    大鼠C6脑胶质瘤血管生成的上升时期在15 d以内,采用该模型进行治疗研究时,干预时间点和观察时段应考虑其本身血管生成的特点。
Abstract:
Objective    To investigate the feature of angiogenesis in C6 brain gliomas by 64-slice MSCT perfusion imaging.     Methods    C6 cells were planted into 40 Wistar rats brain via stereotaxic apparatus. Ten rats were randomly selected at the time periods of 5-9 days, 10-14 days, 15-19 days after cell transplantation to do CT perfusion for the changes of functional cerebral parameters. The data were transferred to the workstation to establish the time-signal curve and the perfusion maps.     Results    The values of cerebral blood flow (CBF) and cerebral blood volume (CBV) increased at 5-9 days, peaked at 10-14 days and maintained stably in a high level. The value of permeability surface (PS) in glioma rats increased at all time periods. No significant difference was found in mean transit time (MTT) of glioma at each time period.     Conclusion    Angiogenesis in the glioma transplanted rats increases in 15 days. The changes of angiogenesis with time must be considered as the factor for therapeutic effect.
