摘要:
目的    建立Wistar大鼠C6胶质瘤模型,采用MRI、CT灌注(CT perfusion, CTP)成像观测肿瘤体积的动态变化,探讨大鼠C6脑胶质瘤的生长规律。    方法    成年Wistar大鼠40只,采用立体定向仪进行C6细胞脑内接种,建立大鼠C6脑胶质瘤模型。每次随机抽取10只接种鼠分别对应于5~9 d、10~14 d、15~19 d 3个时间段行CTP、MRI及病理学检查,观测大鼠C6脑胶质瘤的生长规律,并评价影像学与病理学结果之间的相关性。    结果    15~19 d时间段CTP、MRI观测结果分别与病理学结果比较均有显著性差异(t=3.131, P<0.01;t=2.566, P<0.05),但CTP与MRI观测结果差异无统计学意义(P>0.05)。5~9 d和10~14 d时间段3种技术观测结果比较差异均无统计学意义(P>0.05)。Pearson线性相关分析表明CTP、MRI与病理学三者测量结果间均有显著正相关,病理学与CTP、病理学与MRI、CTP与MRI的t=0.998, 0.998, 1.000(P<0.01)。回归分析表明,肿瘤体积随时间呈指数规律增长(rpatho=0.990, rCTP=0.987, rMRI=0.990, P<0.01)。    结论    影像学活体观测的准确性高,更真实反映肿瘤的实际大小,适于对肿瘤动物模型的生长观测和试验性治疗的疗效评价。
Abstract:
Objective    To establish a Wistar rat model of C6 brain glioma and explore the growth pattern in C6 brain glioma rats by MRI and CT perfusion (CTP) imaging.     Methods    Rat model of brain glioma was established by implanting C6 cells into 40 adult Wistar rats with stereotactic technique. CTP, MRI and pathologic examination were performed in 10 implanted rats randomly sampled at the time periods of 5-9 d, 10-14 d, 15-19 d after tumor implantation. The growth pattern of C6 brain glioma in rats was observed and the correlation between imagings and histopathologic results was evaluated.     Results    There had significant difference at 15-19 d (t=3.131, P<0.01; t=2.566, P<0.05) between CTP, MRI and histopathologic results, but no significant difference (P>0.05) between CTP and MRI; and no significant difference (P>0.05) at 5-9 d, 10-14 d. Pearson linear correlation analysis revealed a significant positive correlation among the growth of tumor shown on CTP and MRI and by pathologic methods (r=0.998, 0.998, 1.000; P<0.01). Regression analysis indicated the tumor volume was in exponential growth with time (rpatho=0.990, rCTP=0.987, rMRI=0.990, P<0.01).     Conclusion    Imaging observation in vivo can accurately reflect the practical tumor volume, and is suitable for the growth pattern of tumor in animal model and to evaluate the efficiency of experimental therapy.
