摘要:
目的    研究大鼠脑出血(intracerebral hemorrhage, ICH)后水通道蛋白9(aquaporin 9,AQP9)在脑组织中的表达变化,初步探讨AQP9在脑出血脑组织病理变化中的作用。    方法    采用定量胶原酶注入大鼠尾状核建立脑出血模型,运用HE染色、免疫组化、免疫荧光双标、RT-PCR等方法检测脑出血大鼠脑组织AQP9表达的变化。    结果    HE染色显示,脑出血面积随着时间的延长而逐渐增大,24 h达到峰值,24 h后无明显变化。RT-PCR结果显示,除ICH 1 h外, ICH早期,AQP9 mRNA含量较假手术组降低,于6 h降至最低;24 h及以后各时段明显增高,于48 h达到峰值,7 d仍明显高于对照。免疫组化和免疫荧光双标显示,AQP9在脑内表达部位随时段变化而变化,其中,ICH 后48 h,AQP9强表达于脑室周围器官和渗透压感受器。    结论    ICH早期AQP9表达降低可延迟细胞内水肿的形成,而ICH后期AQP9表达上调,可能与水电解质平衡调节和能量代谢调节有关。
Abstract:
Objective    To observe the changes of aquaporin 9 (AQP9) expression and its role in rat brain after intracerebral hemorrhage (ICH).     Methods    Collagenase was infused into the right caudate nucleus to establish ICH model in adult Wistar rats. HE staining was used to detect the pathological changes. The protein expression of AQP9 was detected by immunohistochemical method and immunofluorescence, and the content of AQP9 mRNA was detected by RT-PCR.     Results    The area of ICH was gradually increased, and reached the peak at 24 h, then showed no more change. The content of AQP9 mRNA increased in 1 h after ICH, then decreased in 2-6 h and reached the lowest at 6 h, dramatically increased again after 24 h and reached the maximum at 48 h, compared with normal controls and sham operation group. The areas where AQP9 expressed were various in different time periods, especially at 48 h the content of AQP9 in circumventricular organs and osmoreceptor was excessive.     Conclusion    In the early stage of ICH, the decreased expression of AQP9 delays the cytotoxic edema, and in the later stage the increased expression of AQP9 is involved in water-ion homeostasis and energy metabolism.
