摘要:
目的    检测Wnt5a 基因在淋巴细胞系恶性肿瘤患者骨髓中的表达,探讨Wnt5a 基因与淋巴细胞系恶性肿瘤发生的关系。     方法    采用氯化氨试剂裂解红细胞收集有核细胞;半定量RT-PCR方法检测19例淋巴细胞系恶性肿瘤、11例治疗缓解淋巴细胞系恶性肿瘤、27例正常人骨髓标本和10例脐带血CD34+细胞中Wnt5a mRNA的表达;免疫细胞化学法检测部分标本中Wnt5a蛋白表达。    结果    Wnt5a mRNA在正常人、淋巴细胞系恶性肿瘤病例、治疗缓解病例和脐带血CD34+细胞中的阳性表达率分别为88.9%(24/27)、10.5%(2/19)、 72.7%(8/11)、100%(10/10),半定量均值分别为(0.401 5±0.150)、(0.113 9±0.048)、(0.357 0±0.141)、(0.247 5±0.095)。经统计学分析 Wnt5a mRNA在淋巴细胞系恶性肿瘤病例中的表达明显低于正常人(P<0.05);治疗缓解病例与正常人间没有显著性差异(P>0.05),而与未缓解病例间有差异(P<0.05);在脐带血CD34+细胞中的表达低于正常人(P<0.05)但高于淋巴细胞系恶性肿瘤病例(P<0.05);Wnt5a蛋白在正常人中阳性表达,而在淋巴细胞系恶性肿瘤病例中低表达或表达缺失。     结论    Wnt5a基因在淋巴细胞系恶性肿瘤病例中表达缺失而在治疗缓解病例中表达恢复,提示淋巴细胞系恶性肿瘤的发生可能与Wnt5a 基因表达缺失或下调有关,Wnt5a可以作为一个淋巴细胞系恶性肿瘤诊断与疗效评价的指标。
Abstract:
Objective    To detect the expressions of Wnt5a gene in bone marrow from lymphocytic malignant tumor patients and research the relationship between Wnt5a gene and lymphocytic malignant tumor.     Methods    Crack RBC and collect karyocyte with chloride ammonia reagent, and detect the expression of Wnt5a gene using semi-quantitative RT-PCR in 19 cases of lymphocytic malignant tumor, 11 cases of remission of lymphocytic malignant tumor, 27 healthy adults and 10 samples of umbilical cord blood CD34+ cells. The expression of Wnt5a protein was analyzed by immunocytochemical staining in some samples.      Results    The positive rate of Wnt5a mRNA expression in healthy adults, the patients of lymphocytic malignant tumor, the patients in remission of lymphocytic malignant tumor and umbilical cord blood CD34+ cells were 88.9% (24/27), 10.5% (2/19), 72.7% (8/11), 100% (10/10), respectively and their semi-quantitative RT-PCR results were respectively 0.401 5±0.150, 0.113 9±0.048, 0.357 0±0.141 and 0.247 5±0.095. The expression of Wnt5a mRNA was lower in lymphocytic malignant tumor patients and CD34+ cells in umbilical cord blood than that in healthy adults (P<0.05), and that was higher in CD34+ cells in umbilical cord blood than in lymphocytic malignant tumor patients, but no significant difference was found between the patients in remission of lymphocytic malignant tumor and healthy adults (P>0.05). Wnt5a protein expressed in healthy adults, the patients in remission of lymphocytic malignant tumor and CD34+ cells from umbilical cord blood, but not in lymphocytic malignant tumor.     Conclusion    The expression of Wnt5a gene decreased during the development of lymphocytic malignant tumor and restored in the remission of lymphocytic malignant tumor. Our results indicate that lymphocytic malignant tumor may correlate with the decreasing or loss of Wnt5a mRNA expression. Wnt5a gene could be one of the indices for diagnosis and therapy of lymphocytic malignant tumor.
