摘要:
目的    研究铁离子在大鼠实验性慢性脑积水发生中的作用。    方法    SD雌性大鼠75只,采用随机数字表法分为3组:铁离子组、半量铁离子组及人工脑脊液组。分别将浓度为6.67、3.33 g/L的三氯化铁溶液及人工脑脊液各5 μl注入大鼠右侧侧脑室,于术后1、14 d及28 d处死大鼠,评价脑积水发生情况并测量侧脑室侧室指数(侧脑室面积/脑片面积×100),观察脑组织病理变化。    结果    术后28 d(n=10)铁离子组9只大鼠发生脑积水,半量铁离子组(n=5)1只大鼠发生脑积水,两组相比差异显著(P<0.01),人工脑脊液组(n=10)未出现脑积水;铁离子组侧室指数(6.87±2.19)大于半量铁离子组(3.32±0.95)及人工脑脊液组(1.09±0.45)(P<0.01)。铁离子组室周脑组织细胞增生,出现大量泡沫状细胞,脉络丛水肿,含铁血黄素的脑室壁沉积,后期出现脉络丛萎缩,室管膜细胞数量减少,纤毛脱落;半量铁离子组相对较轻;人工脑脊液组无明显变化。      结论    侧脑室注入高浓度铁离子可诱发脑积水。
Abstract:
Objective    To investigate the roles of iron ion in the development of experimental hydrocephalus in rats.     Methods    Seventy-five Sprague-Dawley female rats were divided into 3 groups randomly, that is, the iron group, the semi-iron group and the artificial   cerebrospinal fluid   (CSF) control group. FeCl3 of 5 μl dissolved in artificial CSF with the concentration of 6.67 g/L and 3.33 g/L were slowly infused into the right lateral cerebral ventricle of the rats in the iron group and in the semis-iron group respectively. The rats in the artificial CSF control group were infused 5 μl artificial CSF. The rats were sacrificed 1, 14 and 28 d later to measure the transverse diameter of the lateral ventricle for the evaluation of hydrocephalus and the index of ventricle, and pathology of brain was also observed.     Results    There were significant difference of hydrocephalus between the iron group (9/10) and the semis-iron group(1/5) at 28 d (P<0.01). The index of ventricle in iron group were higher than the semis-iron group and the artificial CSF control group (P<0.01). Cellular proliferation surrounding ventricle, edema of choroid plexus, deposit of haemosiderin, absence of ciliums and microvillares of the ependymal cells were observed in rats of the iron group. The pathological changes in the semis-iron group were milder than in the iron group. The artificial CSF control group showed no hydrocephalus.     Conclusion    Injecting higher concentration of iron into the lateral cerebral ventricle can produce hydrocephalus.
