摘要:
目的    探讨染料木黄酮(Genistein)对人乳腺癌细胞株MDA-MB-453(ER-,HER2-high)、MCF-7/HER2(ER+,HER2-high)、和MCF-7(ER+,HER2-low)增殖能力的影响,并观察HER2在染料木黄酮抑制肿瘤增殖中的作用。    方法    通过基因转染技术建立HER2/neu高表达的人乳腺癌MCF-7细胞,免疫荧光细胞化学法鉴定;不同浓度的Genistein分别处理3种乳腺癌细胞,MTT法分别检测Genistein对肿瘤细胞增殖能力的影响。    结果    Genistein对MDA-MB-453细胞的增殖抑制作用随处理浓度增加而增大,IC50约为100 μmol/L;Genistein对MCF-7/HER2和MCF-7细胞在低浓度表现为增殖促进作用,分别在大于40 μmol/L和10 μmol/L时表现为增殖抑制作用。    结论    Genistein可通过ER依赖和非依赖两种途径,在一定浓度范围内抑制肿瘤细胞增殖,且HER2高表达能够降低染料木黄酮对ER+乳腺癌的增殖抑制作用。
Abstract:
Objective    To investigate the effect of genistein on the proliferation of breast cancer cell lines, MDA-MB-453(ER-, HER2-high), MCF-7(ER+,HER2-low), MCF-7/HER2(ER+ HER2-high), and observe the role of HER2 in the antiproliferative effect of genistein.     Methods    Breast cancer cell line MCF-7 highly expressing HER-2/neu was constructed by cell transfection, and the successful transfection was confirmed by immunofluorescence cytochemical technology. After treated by genistein (5, 10, 40, 70, 100, 130, 160, 190, 220  mol/L) for 48 h, the 3 cell lines were examined by MTT assay to evaluate the antiproliferative effect of genistein.     Results    The inhibitive effect of genistein on MDA-MB-453 was increased along with the increasing concentration of genistein, and the IC50 was 100 μmol/L. The proliferation of MCF-7/HER2 and MCF-7 were enhanced upon a low concentration of genistein, but inhibited at a higher concentration (40 μmol/L and 10 μmol/L, respectively).     Conclusion    Genistein could inhibit the proliferation of all 3 breast cancer cell lines within a certain range of concentration through ER-dependent or ER-independent pathways, and HER2 high-expression could attenuate the antiproliferative effect of genistein on ER positive breast cancer cells.
