摘要:
目的  建立一种稳定、高效、死亡率低的成年大鼠脑室出血后慢性脑积水动物模型。  方法  SD雌性大鼠110只,采用随机数字表法分为3组:生理盐水对照组及侧脑室注血组各50只,正常对照组10只。采用侧脑室注血法于侧脑室注入自体不抗凝血130 μl制备大鼠脑室出血后慢性脑积水动物模型,以术后28 d时大鼠前囟后0.4 mm冠状面的侧脑室宽度评价脑积水的发生情况。分别于术后24 h、72 h、7 d、14 d及28 d处死大鼠,测量上述平面侧脑室指数(侧室面积/脑片面积×100),并观察脑室出血后脑组织病理结构变化。  结果  正常对照组无脑积水,生理盐水对照组在各时相点未出现脑积水,侧脑室注血组28 d时10只大鼠中7只发生脑积水。28 d时侧室指数分别为正常对照组(0.79±0.49)、生理盐水对照组(1.05±0.41)、侧脑室注血组大鼠(4.51±1.94);侧脑室注血组与生理盐水对照组及正常对照组相比均有非常显著差异(P<0.01)。侧脑室注血组大鼠24、72 h脑室周围白质水肿,脑组织细胞增生,胼胝体出现扭曲和肿胀;7 d时侧脑室积血已经消散,出现含铁血黄素侧脑室壁沉积;28 d时室管膜细胞有纤毛和微绒毛的缺失。生理盐水对照组无以上变化。  结论 采用侧脑室注血法可以制备大鼠脑室出血后慢性脑积水模型。
Abstract:
Objective To establish the rat model of chronic hydrocephalus after intraventricular hemorrhage (IVH).   Methods Totally 110 Sprague Dawley female rats were divided into 3 groups: IVH group (n=50), saline control group (n=50) and the normal group (n=10). The rats in IVH group were infused autologous whole blood (130 μl) into the right lateral cerebral ventricle. All rats except the normal group were sacrificed on day 1, 3, 7, 14, 28 after operation, and the transverse diameter of the lateral ventricle was measured to evaluate hydrocephalus and the index of ventricle. The pathological changes of the brain were observed.   Results  Hydrocephalus occurred in 7 out of 10 rats in IVH group at 28 d. The index of ventricle in IVH group (4.51±1.94) was higher than that of saline control group (1.05±0.41) and the normal group (0.79±0.49) (P<0.05). In the brain of the rats in the IVH group, we observed edema of white matter surrounding ventricle, twist and engorge of callosum, proliferation of rhagiocrine cells in 24, 72 h after operation, absence of ciliums and microvillares of the ependymal cells on day 28. No similar changes occurred in the other two groups.   Conclusion Injecting autologous blood into right lateral cerebral ventricle can produce chronic hydrocephalus similar to hydrocephalus after intraventricular hemorrhage.
