摘要:
目的    探讨采用抗坏血酸诱导体外培养小鼠胚胎干细胞(mouse embryonic stem cell, mESC)分化为心肌细胞的可行性。    方法    未分化ES细胞在滋养层上长至汇合,胰酶消化成单个细胞经旋转生物反应器(RCCS)制备拟胚体(embryoid bodies,EBs),EBs贴壁后用抗坏血酸诱导分化为心肌细胞。通过光镜观察跳动的心肌细胞,采用免疫组织化学、激光共聚焦显微镜(CLSM)和RT-PCR的方法鉴定心肌细胞。    结果    抗坏血酸能有效地诱导小鼠胚胎干细胞向心肌细胞的分化,诱导效率为90%;分化的心肌细胞自发而有节律地收缩,心肌肌钙蛋白T(cTnT)、α-辅肌动蛋白(α-actinin)、肌动蛋白(actin)表达阳性。此外,分化的心肌细胞还表达调控心肌发育的基因α-MHC、β-MHC。    结论    抗坏血酸能有效地诱导体外培养小鼠胚胎干细胞分化为心肌细胞。
Abstract:
Objective    To investigate the feasibility of ascorbic acid to induce mouse embryonic stem cells (mESC) to differentiate into cardiac myocytes.      Methods    Undifferentiated mouse embryonic stem cells grew confluent and were digested by trypsin to single cell to form embryoid bodies (EBs) by rotary cell culture system (RCCS). After plated, EBs were induced by ascorbic acid, then observed with light microscopy and confocal laser scanning microscopy (CLSM), identified by immunohistochemistry and RT-PCR.     Results    The cells after induction displayed spontaneous and rhythmic contractile activity. The beating cells was markedly increased in number and accounted for 90% of the induced cells. They were positive for cTnT, α-actinin and actin, and also expressed the cardiac genes α-MHC and β-MHC.      Conclusion    Ascorbic acid could effectively induce mES cells into cardiac myocytes.
