目的 探讨 AngⅡ诱导人血管内皮细胞Ⅱ凋亡的作用及其机制。 方法 原代培养人脐静脉血管内皮细胞,用不同浓度 AngⅡ及神经酰胺合成酶抑制剂烟曲霉素B1(fumonisin B1,FB1)处理细胞,采用 TUNEL法检测细胞凋亡,RT-PCR、Western blot技术对 bax mRNA及蛋白进行表达分析。 结果 AngⅡ 处理组内皮细胞的凋亡阳性率显著高于对照组 (P<0.05)且具有时间和剂量依赖性,对照组和 10-7、10-6、10-5mol/L AngⅡ处理 24 h后各组细胞凋亡率分别为(2.2±1.1)%、(6.0±1.2)%、(17.5±2.3)%、(27.4±4.5)% ;终浓度 10-6 mol/L的 AngⅡ处理 6、24、48 h后细胞凋亡率分别为(6.2±2.3)% 、(15.5±3.1)% 、(21.6±2.5)%; FB1能显著抑制 AngⅡ诱导的内皮细胞凋亡(P<0.05),AngⅡ处理组 bax mRNA及蛋白的表达较对照组显著升高(P<0.05),FB1组 bax mRNA和蛋白表达较对照组没有显著变化(P>0.05)。 结论 AngⅡ通过神经酰胺及 bax诱导细胞凋亡,其中 bax作为神经酰胺的下游基因起作用。
Abstract:
Objective To study the effect of angiotensinⅡ(AngⅡ) on apoptosis in endothelial cells(EC).  Methods Human umbilical endothelial cells were cultured in vitro and treated with AngⅡ in various concentrations alone and in combination with fumonisin B1(FB1, an inhibitor of ceramidase). TUNEL was employed to determine the apoptosis of the cultured EC. The bax mRNA and protein levels of EC were detected by RT-PCR and Westernblot techniques.  Results The results showed that the number of apoptotic cells was significantly higher in AngⅡ-treated endothelium than in the control group(P<0.05). AngⅡ induced EC apoptosis in time- and dose-dependant manner. The percentage of the apoptotic EC were (2.2±1.1)%, (6.0±1.2)%, (17.5±2.3)%, (27.4±4.5)% respectively after treatment with AngⅡ at various concentrations(0, 10-7, 10-6, 10-5 mol/L)for 24 h. Exposing to 10-6 mol/L AngII for 6, 12 and 24 h, the rate of apoptotic cells were (6.2±2.3)%, (15.5±3.1)%, and (21.6±2.5)% respectively. Compared to AngⅡ group, FB1 inhibited EC apoptosis induced by AngⅡ(P<0.05).The expression of bax mRNA and protein in EC treated with AngⅡ significantly increased(P<0.05)but not significant with the FB1 group(P>0.05).  Conclusion AngⅡ may induce apoptosis of endothelial cells via ceramide and up-regulating the bax mRNA and protein expression. Ceramide is the upper stream of bax and induces apoptosis by bax pathway.
