摘要:
目的 观察脉冲电磁场(pulsed electromagnetic fields, PEMF)对5-氮胞苷(5-aza)诱导大鼠骨髓间充质干细胞(mesenchymal stem cells, MSCs)向心肌样细胞分化时细胞增殖的影响。 方法 50 Hz脉冲低频电磁场刺激5-aza诱导后7 d 后的MSCs,根据不同的感应强度分为A、B、C组,每组根据作用时间的不同分为4个亚组;设未暴露PEMFs的细胞为对照组。用MTT法测定细胞增殖变化,用流式细胞技术检测细胞周期。 结果 0.5 mT和1 mT的感应强度下20~30 min细胞比对照组有显著性增殖,5 mT作用30~60 min以及1 mT作用60 min与对照组相比较呈显著性抑制。流式检测细胞周期显示0.5 mT和1 mT在20~30 min内可以引起细胞S+G2%增高,各感应强度下作用60 min以及5 mT感应强度下作用20 min以上均引起S+G2%的下降。 结论 50 Hz脉冲低频电磁场0.5 mT和1 mT的感应强度下,作用20~30 min可通过引起诱导后的MSCs周期中S+G2%增高而促进它的增殖。
Abstract:
Objective〖WTBZ〗To investigate the effects of pulsed electromagnetic fields on the proliferation of rat marrow mesenchymal stem cells induced by 5-azacytidine.  Methods Rat marrow mesenchymal stem cells induced for 7 d with 5-azacytidine were exposed or unexposed to 50 Hz pulsed electromagnetic fields and then randomly divided into A, B, C groups and 1, 2, 3, 4 subgroups according to different inductance intensity and different exposed time. Unexposed under pulsed electromagnetic fields was control groups . Proliferation capacity of rat marrow mesenchymal stem cells induced was examined by MTT method. Cell cycle was detected by flow cytometry.  Results Under 0.5 mT and 1 mT inductance intensity for 20-30 min rat marrow mesenchymal stem cells induced by 5-azacytidine were proliferated significantly in comparison with control groups, while restrained significantly proliferation cell under 0.5 mT and 1 mT inductance intensity for 30-60 min compared with control groups. Cell cycle at S+G2 was increased under 0.5 mT and 1 mT inductance intensity for 20-30 min and decreased under 3 kinds inductance intensity for 60 min and 5 mT over 30 min versus control groups.  Conclusion 50 Hz pulsed electromagnetic field at 0.5 mT and 1 mT inductance intensity for 20-30 min can promote the proliferation of mesenchymal stem cells by increasing cells at S and G2 stage.
