摘要:
目的  观察外源性Wat5a对K562细胞非经典Wnt5a/Ca2+途径的影响,进一步研究外源性Wnt5a诱导K562细胞的分化机制。  方法  激光共聚焦显微镜观察Ca2+内流的变化,以RT-PCR及免疫组化检测β-catenin的表达变化,以Western blot检测cyclin D1的表达变化。  结果  外源性Wnt5a能明显促进K562细胞Ca2+内流,显著下调cyclin D1表达,且能下调K562细胞β-catenin表达,但β-catenin表达下调无统计学意义。  结论  外源性Wnt5a能激活K562细胞非经典Wnt5a/Ca2+途径,外源性Wnt5a诱导K562细胞分化机制可能与此相关。
Abstract:
Objective  To study the mechanism of K562 cell differentiation induced by exogenous Wnt5a in aspect of noncanonical Wnt5a/Ca2+ signaling pathway.   Methods  After K562 cells were cultured in medium supplemented with Wnt5a- or GFP-riched supernatant for 1-5 d, Ca2+ inflow changes were observed by laser confocal microscopy. The expression changes of β-catenin and cyclin D1 were detected by RT-PCR, immunocytochemical staining and Western blotting.   Results  Exogenous Wnt5a obviously promoted the Ca2+ inflow of K562 cells and down-regulated the expression of cyclin D1. Furthermore, the expression of β-catenin was lower, but had no statistically significance.   Conclusion  Exogenous Wnt5a can activate noncanonical Wnt5a/Ca2+ signaling pathway in K562 cells, which may relate to the K562 cell differentiation.
