摘要:
目的 从中药栀子中提取抗内毒素(lipopolysaccharide,LPS)的有效成分并进行活性初步分析。 方法 运用大孔树脂AB-8、聚酰胺树脂、萃取技术及高效液相色谱分离技术对栀子水提物进行分离;通过测定他与lipid A的结合强弱对分离过程进行跟踪,获得粗组分D,经过HPLC技术进一步获得与lipid A有较强结合活性的组分CJ-1;动态比浊法鲎试验检测组分CJ-1在不同浓度(0.5、1、2 mg/L)对LPS(0.2 μg/L)的中和作用;ELISA法检测CJ-1组分对LPS(100 μg/L)刺激RAW264.7细胞释放肿瘤坏死因子-α(TNF-α)的影响。 结果 从中药栀子中逐步分离出与lipid A结合活性较高的组分CJ-1,当CJ-1浓度为0.2 g/L时,RU值为112 arc/s; CJ-1浓度为0.5~2.0 mg/L时,对0.2 μg/L LPS介导的鲎试剂的凝结反应具有显著的抑制作用(P<0.01,P<0.05),并呈剂量效应关系;CJ-1浓度为50~200 mg/L时,对100 μg/L 的LPS介导的RAW264.7细胞释放TNFα具有显著的抑制作用(P<0.01,P<0.05),并呈剂量效应关系。 结论 依靠lipid A为靶点指导中药栀子水煎液分离,从中分离提取出与lipid A的结合作用较强的组分CJ-1,CJ-1在体外对LPS具有一定的中和作用。
Abstract:
Objective To isolate a bio-effective fraction from a traditional Chinese medicine cape jasmine fruit and evaluate its anti-LPS bioactivity.  Methods The fraction, named D and targeting on lipid A (the bioactive component of LPS), was isolated from the water extracts of cape jasmine fruit by using the techniques, such as macroporous adsorptive resins, solvent extraction, polyamide laminar analysis under the screen of an affinity biosensor. Purer components CJ-1 were obtained after high performance liquid chromatography (HPLC). The neutralizing activity to LPS (0.2 μg/L) of CJ-1 at different concentrations (0.5, 1.0, 2.0 mg/L) were further examined by kinetics turbid metric limulus. Enzyme linked immunosorbent assay (ELISA) was also used to detect CJ-1’s effect on LPS-induced release of tumor necrosis factor-α (TNF-α) from RAW 264.7 cells.  Results The isolated components CJ-1 have high binding ability to lipid A (RU value: 112 arc/s). CJ-1 (0.5, 1.0, 2.0 mg/L) inhibit 0.2 μg/L LPS-induced coagulation of limulus reagents significantly and 100 μg/L LPS-induced TNF-α secretion both in a dose-dependent manner (P<0.05).  Conclusion Biosensor technique is effective in guiding the isolation of CJ-1 from water extracts of cape jasmine fruit. CJ-1 has high affinity to lipid A and is able to neutralize LPS in vitro.
