摘要:
目的  研究低氧(2% O2)对成年雄性SD大鼠心脏右心室血管紧张素转换酶2(angiotensin conversion enzyme 2,ACE2)mRNA、蛋白水平表达的影响。  方法  在常氧与低氧环境(模拟海拔5 000 m高原、23 h/d)下饲养SD大鼠,并随机分为缺氧1、3、30 d组和常氧对照组,每组10只,分别在模拟海拔5 000 m高原低压舱内(缺氧各组)和舱外(常氧对照组)测定肺动脉压、右心室功能、左右心室质量指数,采用RT-PCR、Western blot检测各组大鼠心脏右心室心肌ACE2 mRNA、蛋白水平的表达变化。  结果  缺氧组大鼠肺动脉压、右心室收缩压显著高于对照组(P<0.01),缺氧30 d组右心室明显肥大伴右心功能显著上调,同时右心室ACE2 mRNA、蛋白合成均有明显增加(P<0.01)。  结论  慢性低压低氧环境下SD大鼠心脏右心室心肌组织ACE2 mRNA表达、蛋白水平显著上调。
Abstract:
Objective    To investigate the effects of hypoxia on the angiotensin conversion enzyme 2(ACE2) mRNA and protein expressions  in rat right ventricle.     Methods    Forty male adult Sprague Dawley(SD)rats, raised both under normoxia and hypoxia environment (simulating the altitude 5000 meters, 23 h per day) were divided randomly into 4 groups: the control group, the 1-day hypoxia group, the 3-day hypoxia group and the 30-day hypoxia group.  Pulmonary arterial pressure (PAP), right ventricular function, ventricular weight index were detected in all rats.  ACE2 mRNA and protein of right ventricle were measured by RT-PCR and Western blot.     Results    PAP, RVSP were increased significantly in hypoxia groups compared with those in the control group.  Right ventricular hypertrophy and right ventricular function,  ACE2 mRNA and ACE2 protein synthesis in right ventricle were increased accordingly after 30-day exposure to hypoxia (P<0.01).      Conclusion    ACE2 mRNA of the right ventricule and ACE2 protein synthesis were increased under hypoxia environment, suggesting that ACE2 may play a role in hypoxic adaptation of the heart.
