目的〖HTSS〗研究1,25-二羟维生素D3对树突状细胞(dendritic cells, DC)表型和功能的影响及其处理后DC在过敏性气道炎症中的作用。 方法 体外扩增小鼠骨髓来源的树突状细胞,采用流式细胞术和ELISA检测1,25-二羟维生素D3对DC表型和分泌细胞因子IL-10和IL-12的影响;观察1,25-二羟维生素D3处理DC过继卵蛋白致敏小鼠肺组织病理特点和支气管肺泡灌洗液细胞类型。 结果 经1,25-二羟维生素D3处理后,DC表达CD80和 MHCⅡ均明显降低,而CD11c变化不明显;分泌IL-10显著增加,而分泌IL-12显著降低。过继小鼠肺组织未见典型的哮喘样病理改变,支气管肺泡灌洗液细胞类型与空白对照组无显著差异。 结论 1,25-二羟维生素D2可抑制DC成熟,经其处理后DC过继致敏小鼠可诱导对抗原的免疫耐受,避免过敏性气道炎症的发生。
Abstract:
Objective To study the effect of 1,25-Dihydroxyvitamin D3 on the phenotype of dendritic cells (DC) and their function and the role of 1,25-Dihydroxyvitamin D3 treated DC in asthmatic airway inflammation.  Methods DC were harvested by differentiating mouse borrow cells in vitro. The effect of 1,25-Dihydroxyvitamin D3 on phenotype of DC and their secretion of cytokines IL-10 and IL-12 was determined by flow cytometry and ELISA. Pathological changes and bronchoalveolar lavage fluid (BALF) cell types of OVA-sensitized mouse transferred by 1,25-Dihydroxyvitamin D3 treated DC were observed.  Results After DCs were treated by 1,25-Dihydroxyvitamin D3, the expressions of CD80 and MHCⅡ were decreased, whereas the expressions of CD11c were not changed. The secretion of IL-10 was increased, but that of IL-12 was decreased. Typical pathological changes of asthma were not seen in lung tissues of transferred mouse. The BALF cell types were not significantly changed as compared with blank control.  Conclusion 1,25-Dihydroxyvitamin D3inhibits DC maturation. Adoptive transfer of DCs treated with 1,25-Dihydroxyvitamin D3 into OVA-sensitized mouse induces antigen-specific immune tolerance, avoiding the onset of asthmatic airway inflammation.
