摘要:
目的  对大黄素的防治瘢痕的生物学作用进行初步探索,为瘢痕的防治开辟新的途径。 方法  采集我科住院患者的增生性瘢痕标本,进行人增生性瘢痕成纤维细胞培养,分别以不同浓度的大黄素作用于细胞,以MTT法测试药物处理后细胞增殖规律,并以激光共聚焦显微镜观察细胞内[Ca2+]i荧光强度的变化。 结果  在不同浓度组大黄素的处理下,人增生性瘢痕成纤维细胞的增殖呈现出抑制的趋势(P<0.05);细胞内[Ca2+]i的荧光强度也明显上升,其幅度和药物浓度有一定的关联。 结论 大黄素对人增生性瘢痕成纤维细胞有显著的抑制作用,其机制可能和细胞内[Ca2+]i水平的改变有关。
Abstract:
Objective    To investigate the effect of emodin in preventing and treating hypertrophic scar.     Methods    The hypertrophic scar samples were collected from 6 patients admitted to our hospital. The fibroblasts of hypertrophic scar were cultured and treated with emodin (respectively 20, 40, 60 μg/ml) in culture medium. The proliferation of fibroblasts was detected by MTT colorimestric assay and the fluorescence intensity of intracellular free calcium was investgated by confocal laser microscopy.     Results    The proliferation of fibroblasts was inhibited significantly after emodin treatment in a dose-dependent manner, while the fluorescence intensity of intracellular free calcuim was significantly increased.     Conlusion    Emodin can inhibit the proliferation of hypertrophic scar fibroblasts, which mechanism may be related to increasing intracellular calcium
