目的 探讨氨氯地平对人乳腺癌高转移细胞株MDA-MB-231体外侵袭的影响及其相关机制。   方法 用8.34 μmol/L氨氯地平处理肿瘤细胞,以人工重组基底膜侵袭小室(Transwell)观察氨氯地平对MDA-MB-231细胞侵袭的影响;免疫细胞化学方法检测血管内皮生长因子(vascular endothelial growth factor, VEGF)和基质金属蛋白酶-9〖JP2〗(matrix metallo-proteinase,MMP-9)的表达。   结果 8.34 μmol/L的氨氯地平可显著抑制MDA-MB-231细胞的侵袭能力,侵袭抑制率为36.27%;免疫细胞化学检测结果显示,MMP-9和VEGF蛋白的表达在MDA-MB-231细胞中亦明显降低  结论 氨氯地平对MDA-MB-231细胞体外侵袭具有明显的抑制作用,此作用与降低肿瘤细胞的MMP-9和VEGF的表达有关。
Abstract:
Objective To investigate the mechanism that Amlodipine inhibits the invasion of human breast cancer cell line MDA-MB-231.     Methods MDA-MB-231 cells was treated by 8.34 μmol/L Amlodipine for 72 h. The invasion activity of MDA-MB-231 cells was evaluated by using Transwell chambers attached with polycarbonate filters and reconstituted basement membrane (Matrigel). The expressions of MMP-9 and VEGF in MDA-MB-231 cells were examined by immunohistochemistry.    Results Amlodipine significantly inhibited the invasion activity of MDA-MB-231 cells, and the inhibitory rate was (36.27±6.39)%. The expressions of MMP-9 and VEGF in MDA-MB-231 cells decreased obviously by Amlodipine.   Conclusion Amlodipine exhibits the invasion activity of breast cancer cell line MDA-MB-231 and it is related to the reduction of MMP-9 and VEGF expressions.
