摘要:
目的  探讨法尼酯X受体(farnesoid X receptor,FXR)对脂肪酸合酶(fatty acid synthetase, FAS)表达的影响。 方法  用FXR激动剂CDCA作用人肝癌细胞株(HepG2),应用 RT-PCR和Western blotting分别从mRNA水平和蛋白质水平检测FAS的表达情况。 结果  用不同浓度的CDCA(25、50、75 μmol/L)分别作用于HepG2细胞,6、12、24、48 h后,FAS mRNA和蛋白质水平呈时间和剂量依赖性下调。 结论  FXR激动剂可抑制脂肪酸合酶的表达。
Abstract:
Objective    To investigate the effect of the farnesoid X receptor (FXR) on fatty acid synthetase expression.       Methods    Human hepatoblastoma HepG2 cells were treated with a FXR’s agonist, chenodeoxycholic acid (CDCA) at 75 μmol/L for 6, 12, 24 and 48 h respectively, or at 25, 50 and 75 μmol/L for 48 h. Expressions of FAS mRNA and protein were determined by RT-PCR and Western blot analysis respectively.     Results    RT-PCR and Western blot analysis indicated that the mRNA and protein were expressed in a time- and dose-dependent manner in the HepG2 cells treatmented by CDCA.       Conclusion    FXR agonist decreases the expression of fatty acid synthetase.
