摘要: 目的 比较常见的几种贫铀促排剂,寻找高效低毒的药物或组合方案。 方法 分别在给予贫铀溶液前1 h、立即、延迟4 h给予碳酸氢钠、Tiron、dimethylthiourea (DMTU)和diethylenetriaminepentamethylene phosphonic acid (DTPMP)及相应的组合方案,并在3 d后进行肾功能检测及病理学观察。结果提前给药,血清尿素氮和肌酐浓度下降程度显著(P<0.05),下降最明显的是NaHCO3+DMTU组和单用DTPMP组;立即给药,尿素氮和肌酐显著降低的是NaHCO3+DMTU组、单用NaHCO3组和单用Tiron组(P<0.05);延后给药,肾功能均未见明显改善(P>0.05)。提前给药和立即给药,NaHCO3+DMTU组肾脏病变最轻;延后给药,病变比提前给药和立即给药的严重,其中以单用Tiron组肾脏病变最轻。结论 急性贫铀中毒时,提前1 h和立即给药时,NaHCO3+DMTU合用效果较好;延后4 h给药,单用Tiron的效果相对较好。
Abstract:
Abstract: Objective To investigate more effective substances or their combination for depleted uranium (DU) decorporation.  Methods We observed the effects of sodium bicarbonate (NaHCO3),4,5-dihydroxybenzene-1,3-disulphonate (Tiron), diethylenetriaminepenta-methylenephosphonic acid (DTPMP), dimethylthiourea (DMTU) and their combinations for DU decorporation administered 1 h before DU exposure, immediately or 4 h after DU exposure. Three days after DU exposure, the results of renal function test and pathological examination were compared.  Results Large reduction in urea nitrogen and serum creatinine was observed when NaHCO3 combined with DMTU was administered immediately after DU exposure. The most effective treatment tested so far was the combination of NaHCO3and DMTU administered after DU exposure at once. However, the efficacy of the tested substances was only partially effective when they were administered 30 min or longer after DU exposure, among which the efficacy of Tiron after prompt and delayed administration was better than the others.  Conclusion The combination of NaHCO3 and DMTU for DU decorporation is better when administered before or immediately after DU exposure. Tiron for DU decorporation is more effective when administered 4 h after DU exposure.
