摘要:
目的 比较分析肿瘤坏死因子相关凋亡诱导配体(TNF-related apoptosis-inducing ligand, TRAIL)诱导人宫颈癌细胞系HeLa细胞凋亡前后蛋白质组中的差异表达蛋白,以进一步了解TRAIL的作用机制。  方法 以TRAIL诱导人宫颈癌细胞系HeLa细胞凋亡为模型,用双向电泳技术分离HeLa全细胞蛋白质,MALDI-TOF质谱鉴定,MS-FIit数据库分析,RT-PCR半定量方法检测JNK2的表达。  结果 鉴定得到12个明显的差异蛋白点,其中8个蛋白点上调,4个蛋白点下调。JNK2的表达与TRAIL有良好的量效关系。  结论 JNKK、MAPKAPK-2、Dip13 beta、SHPS-1、DDBb、JEM-1、Macoilin、Leucine-rich repeat-containing protein 14、Ena/VASP-like protein、PTPase-MEG2、GRB10 adaptor protein、THUMP domain-containing protein 1为TRAIL诱导HeLa细胞凋亡的新的相关蛋白。
Abstract:
Objective To detect the differential proteins between HeLa cells treated and untreated by TRAIL so as to further understand the mechanisms of apoptosis induced by TRAIL in HeLa cells.   Methods The differential proteins were observed by 2D electrophoresis, identified by matrixassisted laser desorption/ionizationtime of flightmass spectrometry (MALDI-TOF-MS) and analyzed by MS Fit. The expression of JNK2 was examined by semiquantitative RT-PCR method.   Results There were twelve differential proteins between HeLa cells treated and untreated by TRAIL, 8 up-regulated and 4 down-regulated. There was a good dose-effect relationship between JNK2 and TRAIL.   Conclusion JNKK, MAPKAPK-2, Dip13 beta, SHPS-1, DDBb, JEM-1, Macoilin, Leucine-rich repeat-containing protein 14, Ena/VASP-like protein, PTPase-MEG2, GRB10 adaptor protein, THUMP domain-containing protein 1 take part in the apoptosis of HeLa cells induced by TRAIL.
