摘要:
目的  观察促红细胞生成素(EPO)及其受体(EPOR)在急性重型脑外伤后大脑皮质内的动态表达规律,并探讨其意义。  方法  将130只Wistar大鼠分为3组:正常组10只、对照组60只(仅行开颅术)和脑损伤组60只(Feeney法)。分别在伤后5、12、24 h及3、5、7 d断头取脑。采用RT-PCR、免疫荧光法检测EPO和EPOR的mRNA及蛋白的表达。  结果  正常组和对照组各时间点有微量EPO及EPOR的表达,脑损伤组在伤后5 h见少量EPO mRNA和蛋白的表达,12 h和24 h表达升高,至3 d(mRNA 0.691±0.035,免疫阳性细胞78.4±6.32)达到高峰,5 d时下降,7 d时降至正常水平。而EPOR mRNA和蛋白在伤后5 h见少量表达 ,12 h时表达升高,至24 h(mRNA 0.736±0.017,免疫阳性细胞70.2±6.31)达到高峰,3 d(mRNA 0.641±0.027,免疫阳性细胞74.8±5.49)、5 d(mRNA 0.656±0.011,免疫阳性细胞69.5±7.21)和7 d(mRNA 0.771±0.017,免疫阳性细胞70.2±6.59)时仍维持在高表达水平,未见减弱。  结论  大鼠急性重型颅脑损伤后大脑皮质内EPO短暂升高,而EPOR持续高水平表达且呈时间依赖性,提示颅脑损伤后外源性EPO能与持续高表达的EPOR结合产生神经保护作用。
Abstract:
Objective  To investigate the expression of erythropoietin (EPO)and its receptor (EPOR) in adult rat brain after acute traumatic brain injury.    Methods  One hundred and thirty Wistar rats were randomly divided into three groups: normal control group (n=10), sham operation group (n=60) and brain injury group (n=60). The sham operation group and brain injury group were further randomly divided into 6 subgroups respectively according to different time points for sacrifice of the animals (5 h, 12 h, 24 h, 3 d, 5 d, 7 d). The expressions of EPO and EPOR at different time points were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunofluorescence staining.   Results  EPO and its receptor mRNA expressed in normal control group and sham operation group at each time point were just a small amount. RT-PCR and immunofluorescence stain revealed EPO mRNA and protein expressions sharply increased at 12 h, peaked at 3 d, gradually declined to 5 d, returned to normal level at 7 d. EPOR mRNA and protein expressions in brain injury group began to increase at 12 h after injury, peaked at 24 h, sustained to 7 d.   Conclusion  EPOR expression was significantly upregulated at least for 6 days after acute traumatic brain injury, while EPO mRNA and its protein were only shortly elevated. The protective mechanism of EPO on central nerve is possibly related to the markedly increased expression of erythropoietin receptor after traumatic brain injury.
