摘要:
目的    研究丁酸钠和顺铂联合作用对非小细胞性肺癌A549细胞增殖的影响及作用机制。    方法    采用CCK-8细胞计数法测定丁酸钠、顺铂联用对A549细胞增殖活性的影响,流式细胞法检测细胞周期和细胞凋亡的改变,RT-PCR法检测p21基因的表达。    结果    丁酸钠、顺铂联用可抑制A549细胞增殖且呈剂量依赖性,联用后的IC50降低至5.44 μmol/L,与单用DDP的IC50(10.25 μmol/L)相比降低了46.93%(P<0.01)。联用可使细胞周期阻滞于G0/G1期,同时诱导细胞凋亡和p21基因表达的上调。    结论    丁酸钠与顺铂联用能抑制A549细胞生长,其作用机制与促进细胞凋亡及诱导p21基因表达有关。
Abstract:
Objective    To investigate the antiproliferative effect of combination of sodium butyrate (NaB) and cisplatin (DDP) on non-small cell lung cancer cell line A549 and its possible mechanism.     Methods    The antiproliferative effects of combination NaB and DDP were evaluated using Cell Counting Kit-8 (CCK-8) assay. The changes of the cell cycle and the apoptosis were observed by flow cytometry. The mRNA expression level of p21 gene was detected by RT-PCR.     Results    The combination of NaB and DDP could inhibit the growth of A549 cell line in a dose and time dependent manner. The IC50 of the combination group was 5.44 μmol/L and the IC50 of  DDP group was 10.25 μmol/L. The combination of NaB and DDP induced cell cycle arrest at G0/G1 phase, and induced cell apoptosis and up-regulation of mRNA expression of p21 gene.     Conclusion    The combination of NaB and DDP has antiproliferative effects on non-small lung cancer cells, and the mechanism may be related to promoting cell apoptosis and inducing the expression of p21 gene.
