摘要:
目的  观察促肾上腺皮质激素释放激素(corticotropin-releasing hormone, CRH)及其结构相关肽urocortin (Ucn)在大鼠腹腔巨噬细胞的表达及LPS对它们表达的影响。  方法  体外培养Wistar大鼠腹腔巨噬细胞,共收集脂多糖(lipopolysaccharides, LPS)处理0、1、2、4、8 h组细胞,每组含8份分别来自不同大鼠的细胞样本。收集细胞总mRNA(n=8)及总蛋白(n=6),利用RT-PCR及ELISA技术分别检测各组细胞CRH及Ucn mRNA转录水平及胞溶多肽含量。  结果 RT-PCR结果显示,正常大鼠腹腔巨噬细胞中即有一定水平的CRH及Ucn mRNA,LPS刺激后二者水平均显著增高,刺激4 h二者均达峰值,约为对照组的2倍。ELISA结果显示,正常大鼠腹腔巨噬细胞即可表达CRH[(0.07±0.01)ng/107细胞]及Ucn[(1.00±0.48)ng/107细胞]; LPS刺激促进了二者的表达,CRH水平在2 h达高峰[(0.30±0.07)ng/107细胞],Ucn水平在8 h达高峰[(8.68±1.25)ng/107细胞]。  结论  大鼠腹腔巨噬细胞同时表达CRH及Ucn,注适量LPS刺激后,两者表达水平均会显著上调,但Ucn多肽水平显著高于CRH。因此,炎症条件下的巨噬细胞可能是局部CRH家族特别是Ucn的潜在来源细胞。
Abstract:
Objective  To investigate the synthesis of corticotropin releasing hormone (CRH) and urocortin (Ucn), in rat peritoneal macrophages and the effect of LPS on their expressions.   Methods  Peritoneal macrophages were harvested from Wistar rats, and cultured in DMEM with 10% fetal bovine serum.  The total RNA and total protein were extracted from normal control cells and the cells treated by LPS for 1, 2, 4 and 8 h. Each cell group was composed of 8 samples from different rats RT-PCR was used to observe transcriptions of CRH mRNA and Ucn mRNA. ELISA was used to observe the expression of intracelluar CRH and Ucn.   Results  CRH and Ucn mRNA existed in normal rat peritoneal macrophages and LPS up-regulated them to the peak at 4 h. CRH and Ucn peptides also existed in normal rat peritoneal macrophage and they respectively reached the peaks (0.30±0.07) ng/107 cells at 2 h and (8.68±1.25) ng/107 cells at 8 h after LPS treatment.   Conclusion  LPS enhancet the expressions of CRH and Ucn in cultured rat peritoneal macrophages, but Ucn peptide level is higher than CRH peptide. These results indicate the macrophages are the potential resource of CRH and Ucn in inflammatory reaction.
