目的 评价乏氧显像剂18F-硝基咪唑丙醇(18F-FMISO)在肺腺癌动物模型中的诊断价值。 方法 荷肺腺癌T739小鼠经尾静脉注入18F-FMISO后不同时间(30、60、90、120 min)处死,测量18F-FMISO的生物分布,并行PET显像。对照组小鼠在注入18F-氟脱氧葡萄糖(18F-FDG)后行PET显像。 结果 荷瘤鼠肿瘤PET显像清晰。生物分布研究表明肿瘤有明显的放射性摄取。在肾脏和肝脏有较高的放射性分布。肿瘤对血和肌肉的T/NT比值均大于2。 结论 肺部恶性肿瘤组织中18F-FMISO摄取高于正常组织,可通过PET清晰显像,为进一步临床应用于肿瘤的乏氧诊断提供了依据。

Objective To evaluate the diagnosis value of a hypoxic contrast, 18F-fluoromisonidazole (18F-FMISO), in murine model of pulmonary carcinoma. Methods After the injection of 18F-FMISO into the tail, 48 T739 mice bearing the pulmonary adenocarcinoma were sacrificed and divided into 4 groups according different killing times (30, 60, 90 and 120 min). The biodistribution was measured and PET scanning was performed. Another 8 tumor bearing T734 mice served as control, and PET was carried out after the injection of 18F-FDG. Results The tumor PET images with 18F-FMISO were clear. Considerable radioactive uptake on tumor was observed. Higher radioactivity was found in the kidneys and liver. The T/NT ratios of tumor/muscle and tumor/blood were above 2.0. Conclusion The uptake of 18F-FMISO in pulmonary malignant tissues is higher than that in normal tissues, thus the pulmonary neoplasm can be identified accurately with PET imaging. Our preliminary study provides a basis for further clinical hypoxic diagnose of the tumor.
